The method is characterized in that a high performance liquid chromatography is connected with an RID-20A differential refraction detector in series to detect the soluble sugar, glucose and fructose which are both pentose can be distinguished, the effect of accurately detecting three soluble sugars including glucose, fructose and cane sugar at the same time is achieved, the needed sample amount is small, and accurate detection can be conducted with only 0.1 g or even less samples. The method has advantages of few detection steps, simplicity, rapidness, good reproducibility, reduction of experiment errors, improvement of the accuracy of data, simultaneous qualitative and quantitative determination of three soluble sugars comprising glucose, fructose and sucrose, and provision of a scientific basis for accurate determination and rapid detection of mixture of the solublesugars in tomatoes.
