Embodiment 1

[0028] A method for detecting OGN protein expression, said method comprising the following steps:

[0029] A. Preparation of paraffin sections:

[0030] 1) Fixation: Pathological tissues of patients with gastric cancer at different stages (early and advanced) were taken and fixed in 10% neutral formalin overnight.

[0031] 2) Dehydration: Take out the fixed specimens and fully rinse them under running water. After rinsing, dehydrate them step by step from low-concentration alcohol to high-concentration alcohol. 70% ethanol, 120 minutes → 80% ethanol, 120 minutes → 90% ethanol, 120 minutes → 95% ethanol I, 120 minutes → 95% ethanol II, 60 minutes → absolute ethanol, 60 minutes → absolute ethanol II, 60 minute.

[0032] 3) Transparent: a mixture of absolute ethanol and xylene (1:1), 10 minutes → xylene I, 8 minutes → xylene II, 8 minutes.

[0033] 4) Wax immersion: Put the transparent specimen into the melted paraffin for immersion. Wax bath A, 90 minutes → Wax bath B, 60 mi...

Embodiment 2

[0065] A method for detecting OGN protein expression, the detection method is the same as that in Example 1, and the detection object is the pathological tissue of patients with advanced gastric cancer.

[0066] Microscopic examination results such as figure 2 shown. The expression level of OGN in this tissue section is low, and the result is judged as +, without obvious brown-yellow particles, which is a weak positive result of OGN detection. The results indicated that OGN had no or low expression in advanced gastric cancer cells.

[0067] In summary, there are obvious differences in the expression of OGN among patients with different stages, so the use of molecular diagnostic technology is playing an increasingly important role in the clinical practice of individualized treatment of gastric cancer.

Abstract

The invention discloses a method for detecting OGN protein expression and application thereof in gastric cancer auxiliary diagnosis. The method comprises the following steps: taking pathological tissues of a patient with gastric cancer, carrying out paraffin embedding slicing, and dewaxing paraffin sections to water; primarily treating the dewaxed slices, soaking the dewaxed slices in a hydrogen peroxide solution in a dark place, and rinsing the dewaxed slices with pure water; repairing with a citric acid buffer solution and a microwave antigen, cooling to room temperature, rinsing with pure water, putting the slices into a wet box, performing immunohistochemical stroke circle marking, dropwise adding serum derived from a secondary antibody, and sealing; incubating the primary antibody, the secondary antibody and the SABC; carrying out light re-dyeing, dehydration and transparency on hematoxylin, carrying out microscopic examination after sealing, and taking brown yellow or light yellow particles appearing on cytoplasm or cytomembrane as positive expression. The expression condition of OGN in gastric cancer tissue can be detected through immunohistochemical staining of pathologicaltissue of a gastric cancer patient, the method is easy and convenient to operate and accurate in detection, and a basis is provided for diagnosis, treatment and evaluation of the gastric cancer patient.

Description

technical field [0001] The invention relates to a method for detecting OGN protein expression and its application in auxiliary diagnosis of gastric cancer, which belongs to the technical field of biological detection. Background technique [0002] According to the International Agency for Research on Cancer (IARC), gastric cancer (GC) is the fifth most common malignancy in the world. It is estimated that about one million cases of gastric cancer occur each year, and more than half of these cases occur in East Asia, including China, Japan, and South Korea. The proportion of men is significantly higher than that of women. Gastric cancer in men is the third most commonly diagnosed cancer and one of the leading causes of cancer death in less developed countries. In the United States, Europe, and China, because most gastric cancers are diagnosed at an advanced stage, their 5-year survival rate is very low, and the survival rate of gastric cancer patients is relatively low, usua...
