Problems solved by technology

[0005] Although the relationship between GM-CSF factor and pregnancy outcome has been reported, there is no research on the quantitative detection of GM-CSF concentration in embryonic conditioned medium (ECM), and the relationship between GM-CSF in ECM and pregnancy outcome has not yet been reported. However, there are no reports on the use of GM-CSF in ECM as a biomarker for pregnancy outcome prediction or transfer embryo selection

Embodiment 1

[0044] 1) Sample preparation and explanation of specific evaluation criteria:

[0045] In this example, 77 samples were tested by using the above-mentioned method of predicting the success rate of assisted reproductive embryo transfer and selecting embryos for transfer by the concentration of GM-CSF factor, and divided the samples to be tested into high concentration, medium concentration and low concentration according to different concentrations group, and collected 6 blastocyst culture droplets from uncultured embryos under the same conditions as negative control group.

[0046] In this embodiment, there is no significant difference in basic conditions such as age, BMI, duration of infertility, proportion of primary infertility, and endometrium thickness of the subjects corresponding to the samples to be tested.

[0047] In this example, the difference in pregnancy outcomes among different groups was analyzed by the chi-square test in SPSS, and then the relationship between...

Abstract

The invention relates to an application of a GM-CSF factor as a biomarker in screening assisted reproductive transplantation embryos and a method, the method takes the GM-CSF factor as the biomarker, and the concentration of the GM-CSF factor in a blastocyst culture solution of the embryos is in positive correlation with the transplantation success rate of the embryos. The successful transplantation means that the screened embryo can be pregnant after transplantation, and the pregnancy comprises HCG positive, clinical pregnancy and continuous pregnancy. The HCG positive is positive when a pregnancy marker HCG in serum or urine is detected in 11 days after transplantation. The clinical pregnancy refers to intrauterine pregnancy sacs found through vaginal ultrasound 30 days after transplantation, wherein the continuous pregnancy is that the pregnancy lasts for more than 3 months. The method can also be combined with morphological scoring, and the higher the concentration of the GM-CSF factor is, the higher the success rate of embryo transplantation with better morphological scoring quality is.

Description

technical field [0001] The invention relates to the field of assisted reproduction, in particular to the application and method of GM-CSF factor as a biomarker in screening assisted reproductive transplanted embryos. Background technique [0002] Infertility has become a very serious and major social problem at present, and assisted reproductive technology (Assisted Reproductive Technology, ART) is currently the most effective treatment for infertility. Although ART technology is becoming more and more mature, there are still some problems that need to be solved urgently, such as the low utilization rate of eggs, the high rate of multiple pregnancy, and the average number of transplantation cycles. An important reason for these problems is that the current method of selecting embryos for transfer based on embryo morphological scoring in ART technology is relatively single, and there are problems such as strong subjectivity, inconsistent standards, and poor accuracy. Therefo...
