Problems solved by technology

[0004] Coriolopsis belongs to Polyporaceae. Existing studies have found that fungi of this genus mostly contain lignocellulose-degrading enzymes such as laccase, and together with other species of Polyporaceae, play an important role in the material cycle and energy flow of forest ecosystems. Plays an important role, but has not been found to have a role in the symbiotic germination of orchids

Embodiment 1

[0027] 1 Materials and methods

[0028] 1.1 Test strains

[0029] Strain Epi910 was isolated from a protocorm of Magnolias during aseptic germination. In September 2018, tree orchid seeds cultured on OMA medium germinated earlier than other seeds. The fungus was isolated on PDA medium by protocorm tissue section method, and strain Epi910 was obtained after purification, which was transferred to a PDA test tube and stored at 4°C.

[0030] Tulasnella fungus Pap12 (GenBank accession number: MT804628), isolated from the roots of Paphiopedilum sp., can promote the growth of Epidendrum radicans, Cymbidium bicolor and Dendrobium sp.) Seed germination.

[0031] 1.2 Plant material

[0032] The capsules of Sclerophyllum, Shulan and Dendrobium were collected for symbiotic germination.

[0033] Requirements for harvesting capsules: the seeds are mature, the pods are healthy and uncracked, and there are no obvious diseases and insect pests on the surface; after harvesting, wipe the sur...

Embodiment 2

[0052] 1. Symbiotic Germination of Orchidaceae Seeds

[0053] Remove the stalks and residual attachments from the collected capsules, rinse them under tap water for 1 min, absorb excess water with filter paper, and then transfer them to an ultra-clean workbench. Soak and disinfect in 75% ethanol for 30 seconds, rinse with sterile water for 3 times, blot dry with filter paper, cut open the pericarp with a sterile scalpel, pick the seeds with tweezers and spread them evenly on the culture medium.

[0054] The strains Epi910 and Pap12 cultured for 7 days were cut into small pieces and placed in the center of the OMA medium after sowing; at the same time, the seeds were inoculated on OMA medium and MS1 medium without bacteria as controls. 10 dishes were inoculated for each treatment, and 3 capsules were sown for each orchid. After the petri dish was sealed, it was cultured in the culture room, the light cycle was 16h / 8h (light / dark), and the temperature was 25.0±2.0°C. Observe a...

Abstract

The invention provides a Coriolopsis strumosa strain for promoting germination of orchidaceae seeds and application thereof, and belongs to the technical field of symbiotic fungi. The strain Epi910 provided by the invention is derived from fungi in the tissue culture germination process of cymbidium seeds, and is identified as Coriolopsis strumosaby combining sequencing comparison results of fragments such as ITS and nLSU of the strain separated from a culture medium and protocorms with morphological characteristics. The strain Epi910 is colonized in seeds and protocorm cells of cymbidium sinense and cymbidium stipule in a mycelium pellet form, and meanwhile, through a symbiotic germination test, it is verified that the Epi910 has similar functions with mucilage film bacteria, and orchid seeds including cymbidium sinense and cymbidium stipule can be promoted to germinate to form seedlings. The symbiotic germination system of the Coriolopsis strumosa-orchid seeds can be used as a new symbiotic mode except for Tulasnella fungi, and a new research object is provided for comprehensively analyzing the symbiotic mechanism of the fungi- orchid plants.

Description

technical field [0001] The invention belongs to the technical field of symbiotic fungi, and in particular relates to a strain of Ceratozoa magnum that promotes germination of Orchidaceae seeds and an application thereof. Background technique [0002] Orchidaceae plants are one of the more evolved groups of angiosperms. Their seeds are tiny and have no endosperm. They only wrap an immature embryo with a simple seed coat and lack the nutrients required for germination. Under natural conditions, orchid seeds need to establish a symbiotic relationship with fungi, relying on fungi to provide them with nutrients and signal substances to promote germination and seedling formation. The low self-reproduction rate and the destruction of the habitat environment in recent years have led to more endangered orchid resources, and the symbiotic germination of seeds by screening suitable fungi can effectively realize the germplasm conservation of orchids and the ecological restoration of wil...
