Embodiment approach

[0038] The third embodiment of the present invention provides an application of the above preparation method in the preparation of anti-inflammatory drugs.

[0039] The fourth embodiment of the present invention provides an application of iridoid glycosides in Hedyotis diffusa in the preparation of anti-inflammatory drugs.

[0040] In order to enable those skilled in the art to understand the technical solution of the present invention more clearly, the technical solution of the present invention will be described in detail below in conjunction with specific embodiments.

[0041] The reagents used in the following examples are all commercially available.

Embodiment

[0043] 1. Rough extraction of Hedyotis diffusa

[0044] The dried Hedyotis diffusa (200.00 g) was pulverized, extracted with 1 L of 75% ethanol at 80° C. under reflux for 2 h, and repeated three times. Filtrate, combine the filtrates, and evaporate to dryness under reduced pressure at 50°C with a rotary evaporator to obtain 32.31 g of crude extract, which is stored at 4°C for future use.

[0045] 2. HPLC-QTOF-MS / MS Analysis

[0046] HPLC analysis column adopts C 18 Column (250mm×4.6mm i.d., 5μm), the temperature is set at 25°C, using water (A) and acetonitrile (B), adding 0.1% formic acid for gradient elution, the injection volume is 20μL, and the gradient conditions are set as: 15%-20 %B (0-5min), 20%–44%B (5–20min), the flow rate was set at 0.8mL / min.

[0047] A QTOF-MS system with an electrospray ionization (ESI) ion source was used for MS data acquisition. The optimal parameter settings for the identification of iridoid glycosides in the negative ion mode are as follow...

Abstract

The invention discloses a preparation method of iridoid glycoside in oldenlandia diffusa and application of the iridoid glycoside in preparation of anti-inflammatory drugs. A diagnostic ion filtration method is adopted to treat an extract of oldenlandia diffusa, and the appearance time of a chromatogram corresponding to the iridoid glycoside is obtained according to diagnostic ions; and the iridoid glycoside in the oldenlandia diffusa is extracted according to the appearance time by utilizing high-speed counter-current chromatography. According to the method, the iridoid glycoside in the oldenlandia diffusa can be directionally extracted, the extracted iridoid glycoside not only has high purity, but also has anti-inflammatory activity, and meanwhile, tedious sample extraction steps are avoided.

Description

technical field [0001] The invention relates to the extraction of plant active ingredients, in particular to a preparation method of iridoid glycosides in Hedyotis diffusa and its application in the preparation of anti-inflammatory drugs. Background technique [0002] The information disclosed in this background section is only intended to increase the understanding of the general background of the present invention, and is not necessarily taken as an acknowledgment or any form of suggestion that the information constitutes the prior art already known to those skilled in the art. [0003] Natural products have been used in medicine for hundreds of years, and they are good candidates for exogenous exogenous agents (eg, antioxidants, anti-inflammatory agents, antibacterial agents, antineoplastic agents). However, natural products usually contain a large number of compounds, and traditional extraction, isolation, and activity assay methods are time-consuming and laborious, some...
