[0051] Choose healthy, growing branches of pear trees in the same year, clean them with sterile water, wipe and disinfect them with 75% ethanol, clean them with sterile water, and dry them in the air. Use a hole puncher with a diameter of 0.5 cm to punch holes in the branches, and the holes are as deep as the xylem. Endophytic fungal supernatant or conidia (10 8 per mL) to spray the perforated branches. After the treatment, the branches were placed in the incubator to moisturize for 2 days, and each hole was inoculated with a 0.5cm diameter pear tree rot fungus cake, and then soaked with absorbent cotton dipped in sterile water. Wrapped, externally wrapped with a parafilm to moisturize, placed in a light incubator at 25°C for cultivation, measured the length of the lesion after 7 days, and took pictures. Inoculate 10 wells each time and repeat 3 times. Incidence rate = (total vaccination points - asymptomatic vaccination points) / total vaccination points * 100%.

Abstract

The invention relates to the technical field of pear tree rot disease treatment, and discloses a method for preventing and treating rot disease by using endophyte. The method comprises the following steps: S1, preparing penicillium purpurogenum; S2, preparing sterile filtrate; S3, preparing a virus-carrying flat plate; S4, selecting pear tree branches; S5, punching holes in the branches by using a puncher, wherein the hole reaches the xylem; and S6, after treatment, placing the branches in an incubator to be subjected to moisture preservation treatment. According to the invention, a fungus JK2 is separated from a pear tree branch having rot disease, the fungus JK2 is identified as penicillium purpurogenum through morphology and molecular biology, a plate confrontation test shows that the fungus JK2 has strong inhibitory activity on pear tree rot fungi, the inhibition rate reaches 95% or above, an in-vitro pear tree branch identification result shows that the JK2 has a good prevention and treatment effect on the pear tree rot disease, and the penicillium purpurogenum is an endophytic fungus, is harmless to the environment and human bodies, and is beneficial to development of biopesticides.

Description

technical field [0001] The invention relates to the technical field of treatment of pear tree rot, in particular to a method for using endophytic bacteria to prevent and treat rot. Background technique [0002] Pear is one of the most important fruits in my country and is cultivated in various regions. However, in recent years, pears in my country have been threatened by various diseases during the production process, among which rot disease is one of the most serious diseases. Pear tree rot is a fungal disease caused by the pathogen Valsapyri. This disease is relatively common in the main pear producing areas of my country. The affected plants usually show bark rot. dead. At present, chemical control is still one of the most effective means to control fruit tree rot. However, the long-term use of chemical pesticides not only increases the selection pressure of the environment on pathogenic bacteria, but also causes environmental pollution and food safety problems. Theref...
