A test bar is prepared from following steps: preparing gold film electrode on insulating material by using micro electric mechanical system (MEMS); after electrode is treated through surface activation technique, a layer of nano metal grain is assembled on reaction zone on surface of the electrode through method of electrochemical deposition; then, reaction reagent is fixed on the said layer of nano metal grain so as to form reagent layer.
