Embodiment 1

[0022] The method of this embodiment is as follows: wash the rhizomes of turmeric or Dioscorea punctatus, dry them, and crush them to 50 meshes, weigh 200 g of dry powder, use methanol with a volumetric concentration of 60%, and place them in 60 Continuously extract in a water bath at ℃ for 3 hours, 3 times in a row; combine the extracts, centrifuge to separate the precipitate, and distill under reduced pressure to recover all the methanol to obtain the reaction solution 1.

Embodiment 2

[0024] The method of this embodiment is as follows: wash the rhizomes of turmeric or Dioscorea punctatus, dry them, and crush them to 80 meshes, weigh 200 g of the dry powder, use methanol with a volumetric concentration of 80%, and place them at 70 ℃ water bath for continuous extraction for 3 hours, 3 times in a row. The extracts were combined, the precipitate was separated by centrifugation, and all the methanol was recovered by distillation under reduced pressure to obtain a reaction solution 2.

Embodiment 3

[0026] Add water-saturated n-butanol to the extract 1 prepared in Example 1, shake well, let stand, and separate the water phase; add water-saturated n-butanol again, shake well, stand still, separate the water phase and combine twice The extracted n-butanol phase is recovered by distillation under reduced pressure to obtain the crude dioscin extract 1; 0.2000 g of the crude dioscin extract 1 is weighed and dissolved in 200 times the volume of 20% aqueous methanol to obtain a reaction solution 3 .

Abstract

The invention relates to a hydrolysis method of dioscin. A solid acid catalyst is added to the reaction solution containing the dioscin according to the proportion of 0.1g to 100g/100ml of extract and then the reaction solution is placed in water bath of 50 DEG C to 90 DEG C for reaction for 2h to 16h; after the hydrolysis, the reactant obtained is cooled to the room temperature and filtered in vacuum and the filter residue is dried at the temperature of 20 DEG C to 80 DEG C; the filter residue is extracted in organic solution for 4h to 12h continuously, the extract is separated and the organic solution thereof is recycled; finally the dioscin solid is obtained. The solid acid catalyst system adopted by the invention is a multiphase reaction; hydrogen ions are restrained on the surface of the catalyst rather than going into the body of the reaction solution, therefore, the acidification of the reaction solution is avoided and the loss of the hydrogen ions is rather small. The experimental result shows that the pH value of the solution phase after hydrolysis is 4 to 5 and the concentration of the hydrogen ions in the solution is 10,000 times lower than that of the solution which is hydrolyzed by hydrochloric acid, thus greatly reducing the pollution to the environment. The hydrolysis method has simple technology, moderate hydrolysis conditions and high hydrolysis conversion rate of the dioscin which is as high as 98 percent, thus fundamentally solving the problems of serious environmental pollution and equipment corrosion occurring in the inorganic acid catalysis hydrolysis process.

Description

technical field [0001] The invention relates to a method for catalytic hydrolysis of diosgenin, in particular to a method for preparing diosgenin by using a solid acid catalyst to catalyze the hydrolysis of diosgenin. Background technique [0002] Diosgenin (diosgenin), also known as saponin, is extracted and prepared from the rhizomes of plants of the genus Dioscorea (Turmeric Ginger, Dioscorea punctatus, etc.). Diosgenin itself has various pharmacological effects such as hemolysis, hypolipidemic, antibacterial, and anti-inflammatory. In addition, diosgenin is a basic pharmaceutical chemical raw material for the synthesis of steroid hormone drugs and steroidal contraceptives. It can be used for the treatment of rheumatoid arthritis, cardiovascular and cerebrovascular diseases and family planning, etc., and occupies a considerable share in the steroidal drug industry. Important position, with great application value and broad market potential. [0003] The traditional inor...
