Embodiment 1

[0016] Embodiment 1 Sandalwood seed embryo separates sports seedling

[0017] 1. Materials and methods:

[0018] 1.1 Seed collection: From October to December, collect mature (purple-black) fruits of sandalwood trees with a tree age of about 10 years. After removing the outer purple-black fleshy pericarp, wash them with clear water and dry them for later use.

[0019] Preparation of artificial medium: Prepare MS medium according to the literature Narayanaswami S, Norstog K.Plant embryo culture.Bot Rev[J], 1964, 30:587-628, after autoclaving (0.1MPa, 121°C, 15 minutes) Cool to 50°C, press, add 4.8mg gibberellin per 1L of MS medium.

[0020] 1.2 kinds of embryo culture in vitro: peel off the red bony mesocarp of the above-mentioned sandalwood seeds, take out the seeds, soak them in 10% sodium hypochlorite aqueous solution for 15 minutes, rinse them with sterile water for 5 times, and blot the surface dry If there is too much water, inoculate on artificial medium under sterile ...

Embodiment 2

[0027] Embodiment 2 Sandalwood seed embryos use the culture medium of different hormone concentrations to separate sports seedlings

[0028] 1. Materials and methods:

[0029] 1.1 Seed Collection: Same as Example 1

[0030] Preparation of medium: MS medium was prepared and sterilized in the same manner as in Example 1, and prepared by adding 2.0 mg or 6.0 mg gibberellin to each 1 L of MS medium, respectively for test groups 1 and 2.

[0031] 1.2 In vitro culture of embryos:

[0032] Method is with embodiment 1. Peeled seeds were treated with 0.1% mercury chloride (HgCl 2 ) solution for 10 minutes. The processing method after seed disinfection is the same as embodiment 1

[0033] Results: See Table 2

[0034] Table 2 Situation of sandalwood seed embryos in vitro cultured into seedlings

[0035]

Embodiment 3

[0036] Embodiment 3 Sandalwood seed embryos use other hormone medium to separate sports seedlings

[0037] 1. Materials and methods:

[0038] 1.1 Seed Collection: Same as Example 1

[0039] Preparation of medium: prepare MS medium in the same way as in Example 1, add 6-benzylaminopurine (6-BA) in an amount of 2.0mg / L, and then autoclave (0.1MPa, 121°C, 15 minutes ).

[0040] 1.2 In vitro culture of embryos:

[0041] Method is with embodiment 1.

[0042] The results are shown in Table 3

[0043] Table 3 The situation of seedling formation of sandalwood seed embryos cultured on the artificial medium containing 6-BA

Abstract

A rapid in-vitro seedling raising method by utilizing sandalwood seed embryo relates to the method for plant regeneration, in particular to a rapid in-vitro culturing seedling raising method using sandalwood seed embryo. Sandalwood seed embryo is put on an artificial culture media and is aseptically cultured in darkness for ten days at a temperature of 25+/-2 DEG C; on the eleventh day, on conditions that the illumination is increased to 16 hours, the darkness is 8 hours and the illumination intensity is 2000lx, the seed embryo is cultured for 10 days; and then the germinated seedling is transferred into sandy soil to culture for more than 30 days on the conditions that the temperature is 26+/-2 DEG C, the relative humidity is 75+/-5 percent, the illumination intensity is 1500-2000lx and the illumination is 12 hours, and then grows to a germination; 2-6mg of gibberellin or 2.0mg of 6-benzyl aminopurine are added into 1L of MS culture media to form the artificial culture media. The invention has the beneficial effects that the seedling raising time is shortened; the planting percent is high, being up to 87 percentage; the sprout germination is tidy; the mass production and operation are convenient; and the cost is low.

Description

technical field [0001] The invention relates to a method for plant regeneration, in particular to a method for rapidly growing seedlings by in vitro culture of sandalwood seed embryos. technical background [0002] Sandalwood (Santalum album L.) is an evergreen tree of the family Sandalaceae, native to India, Indonesia and Malaysia, and has high medicinal and economic value. Its trunk heartwood is rich in volatile oil, which is a valuable Chinese herbal medicine. It mainly has the effects of warming the stomach, relieving pain and stopping bleeding. The volatile oil contained in the heartwood is widely used in the pharmaceutical industry. In addition, sandalwood is also a valuable raw material in the production of daily cosmetics. Sandalwood is an evergreen plant and is also used as a green tree species in subtropical regions. Therefore, vigorously promoting sandalwood planting is of great significance for solving the shortage of domestic sandalwood resources. [0003] To ...
