Abstract

The invention relates to a preparation and an application of a non-viral gene vector. The non-viral gene vector is a poly-benzimidazole metal complex, which uses poly-benzimidazole TTHB, DTPB, EGTB, EDTB, IDB or NTB as main ligands, uses polybasic carboxylic acid containing benzene rings or aliphatic chains as bridging ligands, uses H2O, Cl-, NO3-, Ac- or ClO4- as auxiliary ligands or counter anions and synthesizes and represents a large amount of complex containing one, two, or a plurality of same and different metal ions of Mg2+, Ca2+, Zn2+, Cu2+, Co2+ / 3+, Mn2+, Fe2+ / 3+ or Ni2+. As novel gene vector, the complex is characterized by simpleness, high efficiency, good water solubility, low cytotoxicity and high transfection efficiency. Experiments of ultraviolet-visible absorption spectrum, light scattering, transmission electron microscopy and the like discover that the complex can effectively induce DNA condensation; and an inverted fluorescence microscope and cell transfection experiments prove that the poly-benzimidazole metal complex can effectively enter cell nucleus for expression as gene released by the vector.
