[0022] 1. Synthesis and purification of superparamagnetic nanoparticles

[0023] Adopt co-precipitation method to synthesize superparamagnetic nanoparticles, specific method is as follows, 324.4mg ferric chloride hexahydrate, 119.3mg ferrous chloride tetrahydrate and 4.5g dextran (Dextran) (molecular weight=10T) are dissolved in 10mL to In deionized water, cool to 2-4°C, add 450 μL 2-4°C ammonia water (25-28%) under strong mechanical stirring and nitrogen protection, and heat the reaction mixture uniformly to 75-85°C within one hour and maintain at this temperature 75 minutes. After the reaction solution was naturally cooled to room temperature, impurities and excess dextran were removed by dialysis (molecular weight cut-off = 300KD), and finally the solution was concentrated by ultrafiltration (300KD).

Abstract

The invention belongs to the technical field of nuclear magnetic resonance, and particularly discloses a magnetic relaxation switch for detecting glycoprotein. A probe of the magnetic relaxation switch adopts a super-paramagnetic nano particle which uses a ferroferric oxide particle as a core, uses glucan as a shell and has average particle size of no more than 60 nanometers. A method for detecting the glycoprotein by using the magnetic relaxation switch comprises two mixing steps and one detecting step, wherein the mixing steps comprises mixing of agglutinin and mixing of target substance, the agglutinin is concanavalin, and the target substance is a1-acid glycoprotein. The linear concentration range of the detection is 0 to 7.0nmol / L, and the detection limit is 0.40nmol / L and is far lower than the normal concentration of AGP in blood plasma.

Description

technical field [0001] The invention relates to the technical field of nuclear magnetic resonance, and specifically relates to a magnetic relaxation switch for glycoprotein detection. Background technique [0002] Glycoproteins are proteins that contain chains of oligosaccharides linked by covalent bonds. Among them, the oligosaccharide chain is linked to the protein by glycosylation in the process of co-translation or post-translation modification. Glycoproteins are found inside cells, in cell membranes and outside cells, and nearly all key molecules in the immune system are glycoproteins. Diversity in protein glycosylation is associated with cell cycle, differentiation and developmental states. Glycosylation at certain sites in proteins is closely related to signal transduction, and abnormal changes in the degree of glycosylation and sugar chain structure are often signs of cancer and other diseases. Some tumor markers and therapeutic targets, such as prostate cancer ma...
