The method for preparation of the flocculant comprises the following steps of: (1) carrying out polymerization reaction on organic carboxylic acid and quarternary-ammonium hexadecyl acrylyloethyl trimethyl aminoethyl ester to obtain a polymeric organic high molecular flocculant; and (2) adding the synthesized polymeric organic high molecular flocculant into polymeric ferric sulfate, heating and stirring to obtain clear transparent light yellow liquid, namely, a phosphorous removal flocculant. The method for application of the flocculant comprises the steps of: adding the prepared flocculant into the wastewater containing phosphorus with the concentration of below 10mg / L, wherein the addition amount of the prepared flocculant is 0.1-0.5mL / L and a pH value is 6.5-8.5; stirring for 30-60min, standing and depositing; and separating mud and water, wherein the content of phosphorus in the wastewater after treatment can be lower than 0.05mg / L.
