Abstract

The method comprises the following steps: (1) conducting domestication and culture on an original seed liquid of microalgae in autotrophic growth conditions by using a mode of increasing the pH value step by step, wherein the initial pH value is 8.0-9.0, which is increased by 2-8 times with pH increasing by 0.5-2 every time; and culturing the microalgae under the same conditions for 24-96 h after each time of pH value increasing, and domesticating and culturing to reach a pH value of 11.0-12.0, so as to obtain a microalgae liquid with high pH tolerance; (2) inoculating the domesticated microalgae liquid to a heterotrophic medium with pH value of 11.0-12.0 and cultivating for 10-24 h, then adjusting the pH vale of the culture medium to 7.0-8.0 and carrying out further culture; and (3) when the microalgae growth achieves a stable period, stopping the cultivation and harvesting microalgae cells.
