Embodiment 1

[0028] The isolation and identification of embodiment 1 bacterial strain

[0029] Take 5g of activated sludge from a sewage treatment plant in Lin'an, add it to 100mL of inorganic salt medium, add 17β-estradiol (E2) to 30mg·L ‐1 As the sole carbon source, at 30°C, 150r·min ‐1 Cultivate on a shaking table for 7 days, transfer to the same medium with 5% inoculum amount, after three consecutive transfers, dilute the enrichment solution in gradient, take 10 ‐4 -10 ‐7 Each 0.1mL of the diluted enrichment solution was applied to add 30mg·L ‐1 On the inorganic salt plate of E2, after incubating at 30°C for 3 days, pick the grown single colony and inoculate it in a medium containing 30 mg·L ‐1 In the inorganic salt liquid medium of E2, at 30°C, 150r·min ‐1 Incubate on a shaking table for 7 days to verify its degradation effect on E2. The formula of inorganic salt medium is: NH 4 NO 3 1.0g·L ‐1 , KH 2 PO 4 0.5g·L ‐1 , K 2 HPO 4 1.5g·L ‐1 , NaCl 1.0g·L ‐1 , MgSO 4 ·7H 2...

Embodiment 2

[0037] Fermentation and immobilization of embodiment 2 restoration bacterial agents

[0038] The process of using the above-mentioned strain JX-2 to produce immobilized restoration bacteria agent is as follows: slant seed—shake flask seed solution—seed tank—immobilized product (the packaging formulation is immobilized bacteria agent).

[0039] 1) Inoculate the Rhodococcus sp. JX-2 test tube species with the preservation number CGMCC 9636 in the fermentation medium, and shake the culture until the logarithmic phase;

[0040] 2) Insert the inoculum amount of the above-mentioned cultured strains into 70% (V / V, based on the volume of the fermenter, based on the volume of the fermenter, The same below), cultivated to the logarithmic phase;

[0041] 3) The seed liquid is inserted into a production tank with a liquid filling capacity of 70% according to 10% inoculum amount for cultivation;

[0042] 4) During the cultivation process of seed tanks and production tanks, the ventilatio...

Embodiment 3

[0045] The biodegradation experiment of bacterial strain JX-2 to 17β-estradiol in the medium of embodiment 3

[0046] In the inorganic salt culture medium (with embodiment 1), add final concentration and be 30mg L ‐1 In E2, strain JX-2 (CGMCC 9636) was inoculated with 5% (V / V, based on the medium volume) inoculum amount, and the inactivated strain JX-2 was used as a control, and cultured in a constant temperature shaker at 30°C. Regularly sampled to detect strain JX-2 at 30mg·L ‐1 The growth of E2 is the only carbon source growth and the degradation of E2, the results are shown in figure 2 . Strain JX‐2 can grow well with E2 as the sole carbon source, and can respond to an initial concentration of 30 mg·L within 7 days ‐1 The degradation rate of E2 reaches more than 90%.

Abstract

The invention discloses a 17 beta-estradiol degrading strain and an application thereof. A rhodococcus sp. JX-2 is preserved in China general microbiological culture collection center at September 4, 2014, and the strain preservation number is CGMCC NO.9636. An immobilized 17 beta-estradiol degrading bacterial agent produced by the rhodococcus sp. JX-2 is provided, a JX-2 bacterial suspension is mixed evenly with a sodium alginate solution, a CaCl2 solution is used as a crosslinking agent, crosslinking is carried out for 5.5-6.5 h, and immobilized small balls with a diameter of about 3-5 mm are obtained. The rhodococcus sp. JX-2 obtained from screening enables the 17 beta-estradiol degradation rate to reach 90% or more under laboratory shake flask culture conditions. The immobilized bacterial agent is applied directly in 17 beta-estradiol polluted water bodies and livestock and poultry manure, and can allow 17 beta-estradiol residues to be reduced by 60% or more in a short period of time.

Description

technical field [0001] The invention belongs to the field of environmental pollution bioremediation, and relates to a 17β-estradiol degrading bacterial strain and application thereof. Background technique [0002] Environmental estrogens (EEs) refer to: entering the organism, they can maintain normal biological organisms by interfering with the synthesis, secretion, transport, binding, active reaction, metabolism, digestion, or producing effects similar to the organism's own hormones. Environmental chemicals that adversely affect the homeostasis, reproduction, growth, and behavior of animals. Estrogen does not need to exist in large quantities to cause strong harm to biological organisms, such as disordering the secretion of sex hormones, reducing the number of sperm, feminizing male animals, increasing the incidence of cancer, etc., causing reproductive, immune, nervous and other systems. Various lesions. A large amount of estrogen is discharged into the environment every...
