Problems solved by technology

However, human collagen expressed by eukaryotic cells still has the problems of difficult purification, low purity and protein degradation
In addition, the amino acid sequence of these collagens expressed by eukaryotic cells is not exactly the same as that of the human body, and there are also defects in biosafety and biocompatibility

Abstract

The invention relates to a protein extract separated from connective tissue and a method and use thereof. The method includes the following steps: transforming a DNA sequence construct (DNA construct) which contains SEQ ID NO:4 or SEQ ID NO:5 to a rat embryo, transplanting the rat embryo to a female rat of the same species and develop to a mature rat, separating a protein extract which contains an objective amino acid sequence from a connective tissue of the mature rat, wherein the objective amino acid sequence (i) is an amino acid sequence which has more than 91% similarity with SEQ ID NO:1 and an amino acid sequence of the SEQ ID NO:3; or (ii) an amino acid sequence which has more than 93% similarity with SEQ ID NO:2 and an amino acid sequence of SEQ ID NO:3; or an amino acid sequence which has more than 91% similarity with SEQ ID NO:1 or an amino acid sequence of SEQ ID NO:3.

Description

technical field [0001] The invention belongs to the field of proteins, and more specifically relates to a protein crude extract separated from connective tissue and its method and use. Background technique [0002] Collagen is a very important protein in animals and an important structural protein in the extracellular space; collagen is also the main component of connective tissue, and also exists in tendons, ligaments, skin, eye corneas and other tissues, accounting for about the total protein in animals 20% to 30% of that. [0003] Collagen is composed of three independent collagen peptide chains, which form hydrogen bonds through glycine on the peptide chains, forming and maintaining a structure of three helical intertwining, called procollagen. After several procollagens are piled up horizontally, aldol condensation reactions will occur between them to form covalent linkages to form collagen microfibrils; several collagen microfibrils undergo similar reactions to produc...
