Embodiment 1

[0061] Example 1 Phenotype identification and genetic analysis of lesion-like mutant lmm9150 of the present invention

[0062] (1) Test materials

[0063] (1) The lesion-like mutant lmm9150 is a lesion-like mutant obtained by screening from the EMS (ethyl methylsulfonate) mutagenesis library constructed by the inventors on the background of Yixiang 1B. The mutant was backcrossed with Yixiang 1B for multiple generations, and the lesion traits of this type were genetically stable.

[0064] (2) Yixiang 1B (indica rice).

[0065] lmm9150 and Yixiang 1B (see figure 1 ) were all from the Genetic Research Laboratory of the Rice Research Institute of Sichuan Agricultural University.

[0066] (2) Test method

[0067] (1) The lesion-like mutants lmm9150 and Yixiang 1B were planted adjacent to each other in the experimental field of Wenjiang Campus of Sichuan Agricultural University. The lesion-like phenotype was observed and photographed at the seedling, tillering and mature stages....

Embodiment 2

[0074] Embodiment 2 Identification test of class lesion mutant lmm9150 of the present invention to bacterial blight resistance

[0075] (1) Test materials

[0076] (1) Lesion-like mutants lmm9150 and Yixiang 1B, bacterial blight-susceptible variety IR24 and bacterial blight-resistant variety IRBB21 were all obtained from the major disease laboratory of the Rice Research Institute of Sichuan Agricultural University.

[0077] (2) Physiological races of bacterial blight of rice: P6, P3, 8428 and Xoo4 were all obtained from the Major Diseases Laboratory of the Rice Research Institute of Sichuan Agricultural University.

[0078] (2) Test method

[0079] (1), the cultivation of rice bacterial blight

[0080] Take Xanthomonas oryzae blight preserved in 50% glycerol, smear evenly on the solid medium, and incubate in the dark at 28° C. for 4 days, and set aside. Described solid medium formula (200ml): sucrose 2g, L-sodium glutamate 0.2g. After mixing sucrose and L-sodium glutamate,...

Embodiment 3

[0088] Example 3 The localization test of the candidate gene of the lesion-like spot mutant lmm9150 of the present invention

[0089] (1) Test materials

[0090] Lesion-like mutant lmm9150, wild-type Yixiang 1B and 02428 (japonica rice) were all from the Genetic Research Laboratory of the Rice Research Institute of Sichuan Agricultural University.

[0091] (2) Test method

[0092] (1) Hybridization of lmm9150 and 02428 to construct F 2 Generation populations were used for genetic mapping. Backcross lmm9150 with Yixiang 1B to construct BC 1 f 2 The generation population was used for MutMap sequencing for gene fine mapping.

Abstract

The invention discloses a rice bacterial leaf blight resistance related gene OsABA2 which consists of nucleotide sequences of SEQ ID No.2 as shown in the specification. The invention further discloses a rice lesion mimic gene which is capable of remarkably improving the resistance of rice to bacterial leaf blight, and a lesion mimic mutant can be acquired through gene edition on the OsABA2 gene. Novel functions of the OsABA2 gene are discovered, and the lesion mimic mutant acquired after the OsABA2 gene is knocked off through gene edition can be applied to rice bacterial leaf blight resistance breeding; meanwhile, the lesion mimic trait is controlled by a single recessive nuclear gene, and hybrid identification and selection can be relatively simple to implement in transgenosis or crossbreeding; and in addition, the lesion mimic gene is derived from nonglutinous rice and has a great promotion function on bacterial leaf blight resistant breeding of the nonglutinous rice.

Description

technical field [0001] The invention belongs to the field of rice genetic engineering, in particular to a rice bacterial blight resistance-related gene OsABA2, and also relates to the use of the gene in regulating rice bacterial blight resistance. Background technique [0002] Rice bacterial blight is a bacterial disease caused by Xanthomonas oryzaepv. oryzae. The main symptoms are yellow-green spots on the leaf tips and edges, which develop into pale and yellow-brown long stripes along the veins, and finally turn grayish white and die. The disease can reduce the yield of rice by 20% to 30%, or 50% to 60% in severe cases, or even fail to harvest. It belongs to one of the three major diseases of rice. Generally, indica rice is heavier than japonica glutinous rice, and late rice is heavier than early rice. Therefore, rice bacterial blight is an urgent problem to be solved in rice production. [0003] Breeding disease-resistant varieties is the most economical and effective m...
