Embodiment Construction

[0052] The following will clearly and completely describe the technical solutions in the embodiments of the present invention with reference to the drawings in the embodiments of the present invention.

[0053] After extensive and in-depth research, the present invention establishes for the first time a molecular imaging mouse model for site-specific knockout of the entire reading frame of the p53 gene and tissue-specific expression of dual fluorescent reporter genes. Specifically, the inventors used homologous recombination-ES targeting technology to convert the translation initiation codon located in the second exon of the mTrp53 gene (GenBank accession number: NM_0116403; Ensembl: ENSMUSG00000059552) located on chromosome 11 of the mouse ATG to knock out the genomic DNA fragment between the translation stop codon TGA located in the 11th exon, and at the same time knock in the Firefly Luciferase-Poly A-CAG-LoxP-Stop-LoxP-mCherry-Poly A gene expression Cassette To the above s...

Abstract

The invention discloses a transgenic construct and application thereof in preparation of a p53 fixed point knock-out and bifluorescence report gene tissue specific expression gene editing mouse. All mTrp53 gene segments on a chromosome 11 of the mouse are knocked out at fixed points, and meanwhile, gene segments with the same length are knocked into the sites, so that the expression of a firefly luciferase gene is regulated by an endogenous p53 gene promoter; the expression of mCherry depends on the tissue specific expression of Cre recombinase. A gene fixed point knock-out / knock-in molecularimage mouse model is prepared for researching a p53 gene transcriptional regulation mechanism; the influence on mouse heredity, development and growth, and tumour occurrence and metastasis caused by p53 gene knockout is monitored in real time noninvasively by utilizing a small animal imaging system, so that the organ selective molecular mechanism of the tumour occurrence is researched, the organ targeting ability of a tumour treatment medicament is observed, and an important model for evaluating tumour medicament treatment pharmacodynamics is established; therefore, the transgenic construct has the application value.

Description

technical field [0001] The invention belongs to the field of genetic engineering, and in particular relates to a transgenic construct and its application in preparing p53 gene entire reading frame site-specific knockout and double reporter gene tissue-specific expression molecular imaging mice. Background technique [0002] Introducing exogenous genes into animal bodies or knocking out endogenous genes in animal bodies or in-depth exploration of gene functions requires transgenic technology, especially to explore the functions of genes in animal bodies or improve biological traits, and to study the role of genes in diseases, especially tumor occurrence and development. and metastasis, requires the use of transgene overexpression or gene knockout techniques. [0003] Disease-related animal models have made great contributions to human health and progress in elucidating gene functions, the pathogenesis of major human diseases, and the development of new drugs. Research on the...
