Abstract

The invention provides a separation and screening method of a trichlorphon degradation strain. The method comprises the following operation steps of (1) soil collection; (2) culture medium preparation; (3) enriched domestication on the trichlorphon degradation strain by a gradient domestication method; (4) trichlorphon degradation strain separation and identification; (5) study on degradation characteristics of the strain on trichlorphon so as to analyze the biological restoration potential of the strain on the trichlorphon polluted soil. The strain screened by the method is obtained by separating in an inorganic salt solid culture medium using the trichlorphon as a unique carbon source; meanwhile, the method can be used for screening other organophosphorus pesticide degrading bacteria; the obtained strain is a soil native microorganism, belongs to a novel strain capable of degrading trichlorphon, and can be applied to the biological restoration of trichlorphon polluted soil. A gas chromatography-mass spectrometry method is used for testing the degradation effect of the strain on the trichlorphon; the operation is simple; the accuracy is high.
