The method comprises the following steps: taking water; filtering impurities, small animals and plants, fish eggs and algae by using filter paper to obtain primary filtered water; filtering microorganisms in the primary filtered water via a microorganism filtering system to obtain secondary filtered water; pouring the secondary filtered water into a one-liter fixed-capacity bottle, and sealing the one-liter fixed-capacity bottle for use; and taking out thesecondary filtered water from the fixed-capacity bottle to perform ultraviolet spectrophotometry of the nitrate. The small organisms and impurities visible to the naked eyes are filtered by the filterpaper, and the microorganisms are filtered by the microorganism filtering system to prevent a change in the nitrate concentration in an inspection process of a water sample; and the content of nitrate is measured by using the ultraviolet spectrophotometry method, an ultraviolet spectrometer is widely used due to the miniaturization and intelligence, and can be operated onsite even if in a farm, and the measurement speed can be greatly improved.
