Embodiment 1

[0034] A method for raising seedlings of gerbera haploid transplanted seedlings, comprising the following steps:

[0035] A. Subculture

[0036] The young shoots of gerbera haploid were excised and transferred to a culture bottle filled with the following proliferation medium for subculture:

[0037] MS medium

[0038]

[0039]

[0040] The subculture conditions of the culture bottle in the culture room are as follows: light intensity 1500-2000lx, temperature 25°C±2°C, light time 10h / d, after 25 days of subculture, transfer to a new container containing the same proliferation culture as above. In the culture bottle of culture medium, and continue to subculture 25d under above-mentioned same subculture condition, repeat like this;

[0041] B. Cultivation of strong seedlings

[0042] When the number of multiplication bases for subculture in step A reaches the required number, transfer to a culture bottle filled with the following strong seedling medium for strong seedli...

Embodiment 2

[0058] Embodiment 2 is the same as Embodiment 1 except for the following operations, and will not be repeated here.

[0059] B. Cultivation of strong seedlings

[0060] In the strong seedling medium: the concentration of kinetin (KT) is 0.05mg / L;

[0061] The light time is 12h / d, and the cultivation time for strong seedlings is 25d;

[0062] C. Rooting culture

[0063] In rooting medium: the concentration of indole butyric acid (IBA) is 0.4mg / L.

Embodiment 3

[0065] Embodiment 3 is the same as Embodiment 1 except for the following operations, and will not be repeated here.

[0066] B. Cultivation of strong seedlings

[0067] In the strong seedling medium: the concentration of kinetin (KT) is 0.1mg / L;

[0068] The light time is 11h / d, and the cultivation time for strong seedlings is 30d;

[0069] C. Rooting culture

[0070] In the rooting medium: the concentration of indole butyric acid (IBA) is 0.5mg / L.

Abstract

The invention discloses a seedling raising method of gerbera jamesonii haploid transplanted seedlings. The method mainly comprises the steps that gerbera jamesonii haploid buds are transferred into aproliferation culture medium for subculture many times, and when the proliferation cardinal number reaches a required number, the gerbera jamesonii haploid buds are sequentially transferred into a strong seedling culture medium and a rooting culture medium for culture; when 4-12 roots grow from base parts of plants, culture bottles are transferred into a greenhouse and stay for 4 days, the sterilized rooting seedlings are transferred into seedling raising bags and grow for 50 days, and the gerbera jamesonii haploid transplanted seedlings are obtained, wherein the greenhouse is covered with a shading net with the shading rate of 70%. After subculture, strong seedling culture is carried out, so that healthy and easy rooting of gerbera jamesonii haploids are promoted; before transplanting, the seedlings are placed in the greenhouse, so that the environment adaptability of the rooting seedlings is enhanced; through seedling selection before transplanting, plant sterilization and timely pesticide spraying after transplanting, and the seedling hardening survival rate and transplanting survival rate of gerbera jamesonii haploid plants are improved. The good-quality transplanted seedlingsare provided for gerbera jamesonii haploid plant cultivation.

Description

technical field [0001] The invention relates to a method for raising seedlings of gerbera haploid transplanted seedlings, and belongs to the field of plant biotechnology and cultivation technology. Background technique [0002] Gerbera is a typical cross-pollination crop. Due to long-term asexual reproduction, it retains high heterozygosity of genotype, and its trait inheritance is extremely complex. In order to make full use of heterosis and eliminate the interference of dominant and recessive, it is necessary to increase the homozygosity of the parents. Conventional breeding methods require multiple generations of selfing to obtain homozygous inbred lines, which takes a long time. Haploid plants are obtained by using pollen (anther) or unpollinated ovary (or ovule) culture, and chromosome doubling becomes a pure line, which can shorten the homozygous breeding time. [0003] Due to the special structure of gerbera flower organs (cross-pollination and stamen abortion durin...
