Problems solved by technology

The emergence of H9N2 subtype AIV was first publicly reported in my country in 1994. At present, this pathogen is widely present in most parts of my country. Co-infection with other pathogens often leads to a serious decline in the production performance of sick poultry and an increase in the mortality rate.

Embodiment 1

[0031] The H9N2 subtype AIV strains TA09 and NDV GM02 were isolated, identified and preserved by the SPF Chicken Research Center of the Institute of Poultry, Shandong Academy of Agricultural Sciences.

[0032] A method for preparing a positive standard for detection of Newcastle disease virus and H9N2 subtype avian influenza virus, comprising the following steps:

[0033] (1) Download the H9N2 subtype avian influenza virus HA gene and NDV F gene reference sequence from GenBank, and use PrimerPremier5.0 to design 2 pairs of full-length amplification primers.

[0034] The amplification primers for the H9N2 subtype avian influenza virus are:

[0035] H9-F: 5'-ATTTCACAACCACTCAAGATGGAA-3';

[0036] H9-R: 5'-AACAAGGGTGTTTTTGCCAATTA-3';

[0037] The amplification primers for NDV are:

[0038] NDV-F: 5′-ATGGGCTCCAAACCTTCTAC-3′;

[0039] NDV-R: 5'-TTGTAGTGGCTCTCATC-3'.

[0040] The expected amplicon size is approximately 1.7 kb.

Abstract

The invention belongs to the field of molecular biology detection, and particularly relates to a positive standard substance for detecting Newcastle disease viruses and H9N2 subtype avian influenza viruses, and a preparation method and application of the positive standard substance. Amplification primers of the H9N2 subtype avian influenza viruses and amplification primers of the Newcastle diseaseviruses are screened, amplified target fragments are connected with vectors, positive recombinant plasmids are screened and verified, the concentration of the positive standard substance is calculated, a standard curve of the standard substance is plotted, and qualitative and quantitative detection is achieved; and finally, by preparing and applying the stable and efficient positive standard substance, qualitative and quantitative detection of samples to be tested is achieved, and thus the positive standard substance is extremely suitable for diagnosis of the Newcastle disease viruses and theH9N2 subtype avian influenza viruses.

Description

technical field [0001] The invention belongs to the field of molecular biology detection, and in particular relates to a positive standard product for detection of Newcastle disease virus and H9N2 subtype avian influenza virus, a preparation method and application thereof. Background technique [0002] Newcastle disease (ND), commonly known as chicken plague, is a highly contagious acute infectious disease caused by Newcastle disease virus (NDV). It is characterized by enteritis, severe diarrhea and neurological symptoms. It is one of the most important diseases and can cause great economic losses. In addition to the first outbreak caused by the pandemic, there have been 3 worldwide epidemics and outbreaks in ND. In the late 1960s, the disease spread rapidly from the Middle East to many parts of the world, causing the second pandemic of the disease. Then, less than a decade later, a third epidemic of the disease occurred in the mid-1970s. The fourth pandemic occurred in t...
