Embodiment Construction

[0025] The invention is a method for determining the content of volatile N-nitrosamine compounds in blood plasma by gas chromatography-mass spectrometry, comprising the following steps:

[0026] (1) Take the blood sample and centrifuge to separate the plasma; take the plasma, add dichloromethane and NDPA-D14 (N-nitrosodi-n-propyl-d 14 -amine) internal standard standard solution, vortex in a vortex mixer, mix well and then ultrasonically extract; centrifuge again after completion; take the lower layer of liquid and inject. Preferably, the blood sample is centrifuged at 3000r / min and 4°C for 2.5-3.5min to separate the plasma. Preferably, calculated by volume fraction, take 1 part of plasma, add 0.45-0.55 part (most preferably 0.5 part) of methylene chloride and 0.045-0.055 part (most preferably 0.05 part) concentration of 500ug / L NDPA-D14 internal standard standard The solution, that is, the volume ratio of plasma, dichloromethane and NDPA-D14 internal standard solution is 1:0....

Abstract

The invention discloses a method for determining the content of volatile N-nitrosamine compounds in plasma by gas chromatography-mass spectrometry. The method comprises the following steps: (1) centrifuging a blood sample to separate out plasma, adding dichloromethane and an NDPA-D14 internal standard solution into the plasma, carrying out whirling in a vortex mixer, mixing uniformly and carryingout ultrasonic extraction, re-centrifuging after the ultrasonic extraction, and feeding lower layer liquid; and (2) carrying out determination by the gas chromatography-mass spectrometry. Through theadoption of the method for determining the content of the volatile N-nitrosamine compounds in the plasma by the gas chromatography-mass spectrometry disclosed by the invention, nine volatile N-nitrosamine compounds in the human plasma can be determined at the same time; the detection limits and quantification limits of the nine volatile N-nitrosamine compounds are 0.01-2.14ng / mL and 0.02-7.14ng / mLrespectively; the study of the load levels of the N-nitrosamine compounds in the human body is facilitated; and a basis is laid for the study of the influence of N-nitrosamine compound exposure on the human health.

Description

technical field [0001] The invention relates to the field of analytical chemistry, in particular to a method for measuring the content of volatile N-nitrosamine compounds in blood plasma. Background technique [0002] Nitrosamine compounds (NAs) are a group of organic compounds with the basic structure R1(R2)N-N=O, which are produced by the reaction of amines or their derivatives with nitrating agents such as nitrous acid, nitrite or nitrogen oxides. It can be absorbed into the human body through the respiratory tract, digestive tract and skin surface. Volatile nitrosamines (VANs) are more toxic than non-volatile nitrosamines (NVANs). Volatile nitrosamines mainly include: N-nitrosodimethylamine (NDMA), N-nitrosomethyl ethylamine (N-nitrosomethyl ethylamine, NMEA), N-nitrosodiethylamine (N -nitrosodiethylamine, NDEA), N-nitrosodipropylamine (N-nitrosodi-n-propylamine, NDPA), N-nitrosodibutylamine (N-nitrosodibutylamine, NDBA), N-nitrosopiperidin (N-nitrosopiperidine, NPIP)...
