A vortex mixer, or vortexer, is a simple device used commonly in laboratories to mix small vials of liquid. It consists of an electric motor with the drive shaft oriented vertically and attached to a cupped rubber piece mounted slightly off-center. As the motor runs the rubber piece oscillates rapidly in a circular motion. When a test tube or other appropriate container is pressed into the rubber cup (or touched to its edge) the motion is transmitted to the liquid inside and a vortex is created. Most vortex mixers are designed with 2 or 4-plate formats, have variable speed settings ranging from 100 to 3,200 rpm, and can be set to run continuously, or to run only when downward pressure is applied to the rubber piece.

The detection method comprises the steps of (1) taking and adding 2 to 4mL of milk into a centrifuge tube, adding 0.3 to 0.5g of demulsifying agent, ultrasonically oscillating for 15 to 30min, then adding 10 to 20mL of ethyl acetate, uniformly dispersing, adding 10 to 30mL of phosphate buffered solution, ultrasonically extracting for 10 to 20min, centrifuging, then transferring 5mL of liquid supernatant, adding 5mL of phosphate buffered solution, uniformly mixing, then dissolving in a sodium hydroxide solution or a potassium hydroxide solution, adding methylbenzene for extraction and taking an upper solution as a standard solution; (2) transferring the standard solution in the step (1) to a solid phase extraction column, leaching by using 5mL of water and 5mL of methanol aqueous solution in sequence, discarding effluent, decompressing and pumping, then eluting with 5mL of methanol and collecting eluant. Blowing to nearly dry with nitrogen at 50 DEG C water bath, adding 5mL of acetonitrile water solution, mixing by a turbine mixer, and obtaining filter liquor through a microfiltration membrane for being detected; (3) adding the filter liquor into a gas chromatograph for detection. The detection method provided by the invention can effectively improve detection result accuracy and provides guarantee for food safety.

The method particularly comprises fatty acid methyl esterification operation and adopts the following method: weighing 0.5 g of an evenly-mixed walnut kernel sample, placing in a 50 mL centrifugal tube with a plug, adding 0.1 mL of 1:1 hydrochloric acid and 10 mL of n-hexane, covering with a lid of the centrifugal tube, carrying out sufficient oscillation for 3 min, carefully opening the lid, releasing gas, centrifuging for 10 min at a speed of 7000 r/min, sucking the supernatant with a rubber-head dropper to a test tube with a plug, and thus obtaining a n-hexane-fat liquid; and taking more than or equal to 2.0 mL of the prepared n-hexane-fat liquid in a 10 mL graduated test tube with a plug, adding 0.8 mL of a 0.5% KOH-methanol solution, carrying out sufficient oscillation for 2 min in a vortex mixer, allowing to stand for 2 min, and sucking the supernatant, namely a fatty acid methyl ester sample liquid. The method is simple to operate, allows the reaction to be carried out only at room temperature, has the advantages of good data repeatability and reproducibility, high precision degree and simple steps, reduces errors caused by incomplete extraction or methyl esterification, internal standard substance preparation and other many aspects, and has fewer error introducing factors.

The method comprises the following steps of: picking up 10-20 capitulums when tubular pyrethrum flowers bloom by 50-99 percent, and placing the capitulums into an oven at 40-80 DEG C for drying for 36-72 hours; pulverizing, then weighing 50-200mg of powder and putting the weighed powder into a glass centrifuge tube; adding 2-10mL of normal hexane, covering the glass centrifuge tube and then oscillating for 20-40s on a vortex mixer; placing the glass centrifuge tube in an ultrasonic water bath for 5-15min, taking out and then continuing to oscillating for 20-40s; centrifuging for 3-8min at a speed of 1,500-3,000 turns / min at room temperature of 20-25 DEG C; absorbing 0.1-0.5mL supernate, and metered by the volume, diluting for 40-80 times with absolute ethyl alcohol as a solution to be measured; measuring the light absorbing valve by an ultraviolet and visible spectrophotometer under the condition of a wavelength of 225nm; and calculating the content of the pyrethrin. The invention has the advantages of simple pretreatment, low testing cost, high speed and good accuracy and can improve the field breeding measuring efficiency and the selecting efficiency of good genotypes.
