[0051] 1.1 Selection of viral proteins

[0052] Based on existing studies, ASFV attenuated infection can resist the same type of strong infection, but cannot resist different types of strong infection. Therefore, in order to find the specific CTL expression for the Chinese strain ASFV China / 2018 / AnhuiXCGQ (GenBank: MK128995.1) At the same time, the protein difference analysis between different genotype strains of type I / II was carried out, and the protein and structural protein with large variation were screened. The specific steps are as follows:

[0053] (1) According to the research of scientists such as Linda Dixon, the following four different genotype strains were selected for analysis and comparison, see figure 1 :

Abstract

The invention provides an African swine fever cytotoxic T lymphocyte (CTL) epitope polypeptide and an application thereof, and belongs to the technical field of preparation of polypeptide vaccines. According to the invention, polypeptides which are derived from African swine fever virus (ASFV) and can be stably bound by major histocompatibility complex class I (MHC I) are screened through a bioinformatic method and a method for identifying binding motifs of MHC I, and the screened polypeptide has the capability of starting CD8+T cells to generate immune response and stimulating organisms to specifically generate CTL, wherein an amino acid sequence of the polypeptide is shown in any one of from SEQ ID NO.1 to SEQ ID NO.18. The polypeptide provided by the invention can be used for preparingAfrican swine fever specific epitope vaccines or polypeptide vaccines, has high safety and good specificity, and has a potential application value in prevention and control work of African swine fever.

Description

technical field [0001] The invention belongs to the technical field of polypeptide vaccine preparation, and in particular relates to African swine fever CTL epitope polypeptide and application thereof. Background technique [0002] African swine fever (ASF) is an acute, hemorrhagic, severe infectious disease caused by African swine fever virus (ASFV) infecting domestic pigs and various wild boars (African wild boar, European wild boar, etc.) . It is characterized by a short onset course, generally 5-14 days, and the most acute and acute infection mortality rate is as high as 100%. African swine fever is caused by a double-stranded DNA virus, the only member of the African swine fever virus family. The serotype is classified based on the hematocyte adsorption inhibition test (HAI), and 32 ASFV strains can be divided into 8 serogroups; the genotype is determined based on the single gene sequence encoding the VP-72 protein, which is divided into 24 genotypes . The genome si...
