Embodiment 1

[0026] A method for removing endotoxin in pectin provided by the invention comprises the following steps in turn:

[0027] (1) Dissolving pectin

[0028] 20 g of pectin was weighed and dissolved in 1 L of water under stirring conditions, the mass concentration of pectin was 2.0%, the stirring speed was 1,500 rpm, the temperature was 30° C., and the stirring time was 2 h.

[0029] (2) Microfiltration of pectin solution

[0030] The completely dissolved pectin solution is sequentially filtered through microporous membranes with a pore size of 0.8, 0.45 and 0.22 μm. The filtration temperature is 70° C. and the pH value of the solution is 4 to remove particles with a particle size larger than 0.22 μm.

[0031] (3) Gel cleaning

[0032] Add 1M HCl solution to the filtered pectin solution to adjust the pH to 1.0 to form an acid gel, and stir to break the gel. The stirring and washing time is 2 hours, the temperature is 30°C, and the number of washings is 3 times. After cleaning, ...

Embodiment 2

[0039] A kind of method for removing endotoxin in pectin provided by the invention, comprises the following steps successively:

Abstract

The invention discloses a method for removing endotoxin in pectin. A purpose of the invention is to provide an endotoxin removing method with characteristics of simple operation, high product yield and easy industrial production. The preparation method is characterized by comprising the following steps: (1) dissolving pectin in water to form a solution; (2) carrying out microfiltration separationon the pectin solution to remove solid impurities; (3) adding an acid solution into the pectin solution to adjust the pH value to form an acid gel, stirring to smash the gel, cleaning, carrying out reduced pressure suction filtration to remove moisture, adding an alkaline solution to adjust the pH value, and dissolving the gel to form a concentrated solution; (4) adding absolute ethyl alcohol intothe pectin solution, and precipitating; and (5) drying the pectin precipitate at normal pressure.

Description

technical field [0001] The technology relates to the field of biopharmaceuticals, in particular to a method for removing endotoxin in pectin. Background technique [0002] Pectin is a natural polysaccharide polymer extracted from plant fruit. It is a mixture of oligosaccharides and polysaccharides with common characteristics. Its main component is D-galacturonic acid, except for galacturonic acid , also contains some neutral sugar components, such as rhamnose, arabinose and galactose. The molecular weight of pectin is between 50,000 and 300,000. It is a white to reddish-brown powder. It is colorless, odorless, has no fixed melting point and solubility, and is insoluble in organic solvents such as ethanol. It is soluble in hot water and slightly soluble in cold water. Pectin is a safe and non-toxic natural food additive recommended by the FAO / WHO Joint Committee on Food Additives, with no daily limit. Pectin has many functions and can be widely used in the food industry as ...
