Abstract

The invention discloses a poplar Melampsora laricipopulina single spore propagation method, and belongs to the technical field of microbial cultivation. The method comprises the steps of using a 90 mm disposable culture vessel; padding two pieces of filter paper at the bottom of the culture vessel; adding 3 mL of sterile water for moisture preservation; dispersing single sori on infected leaves into4 [mu]L of an agar water solution; spraying a solution mixed uredinium onto tender leaves of NL895 with the disease infection capability; after inoculation is completed, putting the materials into agreenhouse with the temperature of 19 DEG C and the light period of 16 h / 24 h to be cultured; after the morbidity, performing further propagation to the sufficient quantity; and performing storage ina refrigerator of 4 DEG C for use.
