Problems solved by technology

[0003] The production process of most existing viral vaccines is based on the adherent cell platform. Typical culture systems such as roller bottles, multi-layer cell culture dishes and microcarrier bioreactors have certain limitations when scaling up. The pipeline in the device is easy to bend and twist, causing the pipeline to be easily damaged. The connection between the pipeline and the main tank in the device is easily caused by twisting, resulting in insufficient sealing, causing the pipeline to loosen, and affecting the culture results. With continuous perfusion, the liquid enters and exits at the same time, and the virus that has not entered the cell in the tank will gradually be discharged out of the tank, which is inefficient and affects the use of the device.

Embodiment 1

[0022] Embodiment 1: as Figure 1-3 As shown, a fixed-bed bioreactor for virus vector vaccine cultivation of the present invention and its use method include a main tank body 1, a circulating perfusion bottle 7, a feed bottle 9 and a harvest bottle 14, and the top of the main tank body 1 is fixedly connected with The tops of the two stoppers 2, the circulating perfusion bottle 7, the feeding bottle 9 and the harvest bottle 14 are all fixedly connected with a fixed block 12, which is convenient for fixing the pipeline and prevents the pipeline from being damaged.

[0023] The cross-section of the limit block 2 is an "I"-shaped structure, which is convenient to support on the limit block 2, so that the end of the limit block 2 close to the main tank body 1 will not bend, preventing the pipeline from being damaged due to multiple bending. 2. The upper thread is connected with a threaded pipe 3. The bottom of the threaded pipe 3 passes through the limit block. 3. One end is fixed...

Abstract

The invention provides a fixed bed bioreactor for viral vector vaccine culture and a use method thereof. The fixed bed bioreactor comprises a main tank body, a circulating perfusion bottle, a material supplementing bottle and a harvesting bottle, wherein the top of the main tank body is fixedly connected with two limiting blocks; and the tops of the circulating perfusion bottle, the material supplementing bottle and the harvesting bottle are fixedly connected with fixing blocks. The fixed bed bioreactor has the beneficial effects that the limiting blocks are used for preventing the looseness and twisting of a pipeline, so that the untight sealing is avoided; the fixing blocks are used, so that the angle of the pipeline is conveniently maintained, and the pipeline is prevented from bending; and a rotary type non-woven polyethylene terephthalate (PET) fixed bed is used, so that the fixing of cells is convenient, and the cells are prevented from flowing out of a reaction tank; by using the combined type of continuous perfusion and circulating perfusion, the virus infection efficiency is improved.

Description

technical field [0001] The invention relates to the technical field of vaccine production equipment, in particular to a fixed-bed bioreactor for cultivating virus vector vaccines and a method for using the same. Background technique [0002] Virus vaccine refers to a kind of biological agent that produces autoimmunity and prevents diseases after the human body is inoculated. Fixed bed bioreactor is an important process for cultivating virus vaccines. The connection between the pipeline and the main tank in the device is easily caused by twisting, resulting in insufficient sealing, causing the pipeline to loosen, and affecting the...
