Embodiment 1

[0036] Recombinant Human Angiostatin K1-3

[0037] Material:

[0038] Egg source: local chicken farm; recombinant human angiostatin K1-3;

[0039] step:

[0040] 1. Egg preparation: Disinfect the surface of the egg with 75% alcohol, and after drying, put the egg with the air chamber upward and the long axis at 45° to the egg tray and put it in an incubator at 37.8°C and 60% relative humidity.

[0041] 2. Egg hatching: During the hatching process, turn the eggs every 8 hours, and the angle of turning the eggs should be 45 degrees in the horizontal position. When hatching to 2.5 days, use 75% alcohol to sterilize the shell of the egg in an ultra-clean bench, lightly knock a crack in the middle of the egg, quickly break the egg shell (to prevent the egg shell from scratching the yolk), and pour the chicken embryo into a sterile in a glass beaker. Due to gravity and the fixation of the egg frenulum, the embryonic disc of the chicken egg can be automatically turned over to the ...

[0046] endostatin

[0047] Material

[0048] Egg source: local chicken farm; endostatin;

[0049] step:

[0050] 1. Egg preparation: Disinfect the surface of the eggs with 75% alcohol, and after drying, place the eggs with the air chamber upward and the long axis at 45° to the egg tray into an incubator at 37.8°C and 60% relative humidity.

[0051]2. Due to gravity and the fixation of the egg frenulum, the embryonic disc of the chicken egg can be automatically turned over to the surface of the yolk. After the beaker is covered, it is...

[0056] recombinant human vascular endothelial growth factor

[0057] Material

[0058] Egg source: local chicken farm; recombinant human vascular endothelial growth factor (purchased from R&D SYSTEM, USA).

[0059] step:

[0060] 1. Egg preparation: Disinfect the surface of the eggs with 75% alcohol, and after drying, place the eggs with the air chamber upward and the long axis at 45° to the egg tray into an incubator at 37.8°C and 60% relative humidity.

Abstract

The invention relates to a chicken germ yolk capsular vessel generating model, as living test model for detecting or selecting the drug which can affect novel vessel. The invention comprises that removing frame of 60h old germ to be put into a small beaker to cultivate for 12h, adding object drug into a silica gel ring on the yolk capsule, the contrast liquid and object liquid both contain glycerin or dimethyl sulfoxide as penetrant, taking off the silica gel ring after 12h, dynamically watching the effect of drug on the yolk capsular vessel. The function of drug on the vessel as restrain, simulate or non function can be judged according to the growth state of the yolk capsular vessel. Compared with traditional model, the invention is simple, visual, quick and accurate.

Description

technical field [0001] The invention relates to the establishment of a new type of angiogenesis in vivo model, provides a chicken embryo yolk sac membrane angiogenesis model, and discloses the use of the model in detecting and screening drugs that affect angiogenesis, and belongs to the field of biotechnology. Background technique [0002] After Folkman of Harvard Medical School proposed the anti-angiogenesis theory in 1971, great progress has been made in the research on the mechanism of angiogenesis, the screening of endogenous angiogenesis stimulators and inhibitors, and the screening of anti-angiogenesis active drugs. Research on angiogenesis mainly relies on experimental methods and models for vascular endothelial cell proliferation and angiogenesis in vivo and in vitro. Among them, the qualitative and quantitative studies on the influence of angiogenesis in vivo use experimental models such as rabbit or rat corneal pouch, mouse back pouch, hamster buccal pouch, mouse e...
