Problems solved by technology

[0006] The technical task proposed by the present invention and the technical problem to be solved are to overcome the translucent, moist, cohesive embryogenic callus induced by the existing Pseudomonas off-vitro culture technology. Loss of the ability to differentiate green shoots, and high seed heterogeneity, not suitable for micropropagation, screening of somatic cell mutants and Agrobacterium-mediated gene transformation and other defects

[0032] 1. Materials and Methods

[0033] 1.1 Experimental materials

[0034] Take the stem section of Pseudomonas spp. with lateral buds as the explant material.

[0035] 1.2 Medium

[0036] The basic medium is composed of macroelements, trace elements and vitamins of MS, 30g / L of glucose and 7.5g / L of solidifying agent-agar are added. MS medium is the most commonly used medium in plant tissue culture (Murashige T, Skoog F.A revised medium from rapid growth and bioassays with tobacco tissueculture[J].Physiol.plant, 1962,15:473~497);

[0037] Lateral bud growth medium: basic medium supplemented with hormone BAP (China Pharmaceutical Shanghai Chemical Reagent Company) 2.0mg / L, NAA (Nanjing Shengxing Biotechnology Co., Ltd.) 0.8mg / L;

[0038] Callus induction and subculture medium: add hormone 2,4-D (China Pharmaceutical Shanghai Chemical Reagent Company) 1.0mg / L and BAP 0.1mg / L to the basic medium;

Abstract

The invention relates to a centipede grass lateral bud inducting callus and plant regenerating method, which belongs to the technical field of the plant tissue culture. Lateral buds of a centipede grass are used as explantation material and vaccinated on callus inducting culture mediums of 2, 4-D-dichlorophe noxyacetic acid 1.0mg / L and BAP-6- benzyl aminopurine, and the inducting frequency of thecallus reaches more than 90%. The callus inducting culture mediums are transferred to a green seedling differentiation culture medium which is attached to a KT-sub- activator after being cultivated twice, and the green seedling differentiation reaches more than 12%. Lateral sprouts are used as explantation material, and yellow, compact and granular callus which is achieved is applicable to centipede grass micro-propagation, micro-propagation, somatic cell cultivation and agrobacterium mediating method gene conversion. The higher the employing quality of lawns is, the lower the quality of seedsis, and using lateral sprouts as explantation has special and important application value to establishment of grasses in lawns regeneration system, improvement of warm-season type grasses in lawns and industrialization of new species.

Description

1. Technical field [0001] The invention relates to a technique for inducing callus tissue and plant regeneration from the lateral buds of Pseudomonas spp., which belongs to the field of plant tissue culture, and is suitable for the cultivation technology of micropropagation, somatic cell culture and Agrobacterium-mediated gene transformation of Pseudomonas spp. 2. Technical Background [0002] Eremochloa ophiuroides (Munro.) Hack. is a perennial herbaceous plant of the Poaceae Ophiuroides genus, and is the only species in the genus Ophiuroides that can be used as turfgrass (Bouton et al, 1983; Hanna, 1995). It has strong stolons, strong and rapid spreading, slanted stalks, beautiful leaf shape, low plant, low maintenance level, barren resistance, less pests and diseases, and can be widely used in garden lawns, rest lawns and soil and water conservation lawns. in (Hanson et al., 1969; Beard, 1973). [0003] Breeding new varieties by conventional hybridization technology not ...
