Embodiment Construction

[0015] In order to better understand the present invention, the present invention will be further described below in conjunction with the implementation examples.

[0016] This embodiment adopts the enteric coating carrier material Eudragit L-100 (Eudragit L-100) produced by German Rohm company.  L-100, produced by Röhm GmbH & Co.KG, Darmstadt, Germany, is a copolymer of methacrylic acid and methyl methacrylate (1:1). The product standard complies with: USP / NF / Ph.Eur.Shenzhen Youpuhui Industrial Development Co., Ltd.).

[0017] Disperse 1 g of enteric-coated Eudragit L-100 evenly in 0.1M acetic acid-sodium acetate buffer solution with a pH value of 5.5, and configure it as a 2% carrier solution; add 4 mL of cellulase solution with a concentration of 15 mg / mL and 0.3 g The cross-linking agent EDC was slowly stirred at room temperature for 3-6 hours; the pH value was adjusted to 3.8 with 3M acetic acid, and centrifuged at 5000 rpm for 10 min; the precipitate was dissolved in ...

Abstract

The invention provides a preparation method for an immobilized cellulase of an enteric coating material, which comprises the following steps: 1) evenly dispersing the enteric coating material Eudragit L-100 into the acetic acid-sodium acetate buffer solution with a pH value of 5.5; 2) adding in the cellulase solution and crosslinking agent solved into the acetic acid-sodium acetate buffer solution with the pH value of 5.5 in sequence, and agitating and stewing for 3 to 6 hours under room temperature; 3) using 3M acetic acid to adjust the pH into 3.8, and centrifuging the solution for 10 min at the speed of 5000rpm; 4) then precipitating and dissolving the enteric coating material into the acetic acid-sodium acetate buffer solution with the pH value of 5.8, thus obtaining the immobilized cellulase. By changing the pH value of the solution, the immobilized cellulase can conveniently complete the homogeneous enzymolysis and heterogeneous recovery of the immobilized enzyme, and is able to ensure that the immobilized enzyme conducts high efficiency enzymolysis of the insoluble cellulose material in homogeneous state, and the by-product lignin after the enzymolysis can be quickly separated. Therefore, the immobilized cellulase of an enteric coating material has the advantages of capacity for repeated use, minor change in the enzymatic activity retention rate, simple and convenient preparation process, much lower costs of using the cellulase, and good popularization and application values.

Description

technical field [0001] The invention relates to enzyme immobilization technology, in particular to a preparation method of enteric coating material immobilized cellulase. Background technique [0002] Cellulose is one of the most abundant renewable resources in nature. Cellulase is used to saccharify it and then ferment it to produce ethanol. Pure ethanol or mixed with gasoline can directly replace gasoline, diesel and other fossil fuels, which can effectively alleviate human fossil energy consumption. growing shortages. However, in the process of hydrolyzing cellulose, cellulase has poor stability, short service life, and low catalytic efficiency, which leads to high production costs of enzymes in the saccharification process of cellulose, which affects its industrial application. Therefore, how to reduce the production cost of cellulase and increase its stability and service life is the key to open up the technical route from cellulosic raw materials to fuel ethanol, and ...
