Embodiment 1

[0016] 1. Steam-exploded straw

[0017] Add 150g of straw cut to a length of 10-15cm, add 150ml of water, stir evenly, put it into a steam explosion tank, and feed steam. After the cold air in the tank is discharged, close the charging valve, increase the pressure to 1.0MPa, and maintain the pressure for 10 minutes. Open the discharge valve, instantly decompress and release the steam-exploded straw, and air-dry it for later use.

[0018] 2. Enzymatic hydrolysis of steam-exploded rice straw

[0019] Take 2g of steam-exploded rice straw, 30IU FPA / g substrate of cellulase and 1IU / g substrate of ferulic acid esterase in the system, then supplement the enzymatic hydrolysis system to 50ml with pH4.8 acetate buffer, and place in a constant temperature water bath at 50°C Medium enzymatic hydrolysis for 6h. Sampling was carried out, and the content of ferulic acid in the sample was determined by HPLC.

Embodiment 2

[0021] 1. Steam-exploded straw

[0022] Add 150g of straw cut to a length of 10-15cm, add 150ml of water, stir evenly, put it into a steam explosion tank, and feed steam. After the cold air in the tank is discharged, close the filling valve, increase the pressure to 1.5MPa, and maintain the pressure for 4 minutes. Open the discharge valve, instantly decompress and release the steam-exploded straw, and air-dry it for later use.

[0023] 2. Enzymatic hydrolysis of steam-exploded rice straw

[0024] Take 2g of steam-exploded rice straw, 30IU FPA / g substrate of cellulase and 1IU / g substrate of ferulic acid esterase in the system, then supplement the enzymatic hydrolysis system to 50ml with pH4.8 acetate buffer, and place in a constant temperature water bath at 50°C Medium enzymatic hydrolysis for 8h. Sampling was carried out, and the content of ferulic acid in the sample was determined by HPLC.

Embodiment 3

[0026] 1. Steam-exploded straw

[0027] Add 150g of straw cut to a length of 10-15cm, add 150ml of water, stir evenly, put it into a steam explosion tank, and feed steam. After the cold air in the tank is discharged, close the filling valve, increase the pressure to 1.6MPa, and maintain the pressure for 4 minutes. Open the discharge valve, instantly decompress and release the steam-exploded straw, and air-dry it for later use.

[0028] 2. Enzymatic hydrolysis of steam-exploded rice straw

[0029] Take 2g of steam-exploded rice straw, 20IU FPA / g substrate of cellulase and 1IU / g substrate of ferulic acid esterase in the system, then supplement the enzymatic hydrolysis system to 40ml with pH4.8 acetate buffer, and place in a constant temperature water bath at 50°C Medium enzymatic hydrolysis for 8h. Sampling was carried out, and the content of ferulic acid in the sample was determined by HPLC.

Abstract

The invention belongs to the application field of biotechnology and particularly relates to a method for improving the extraction rate of ferulic acid in plant straws by steam explosion pretreatment. The method includes the following steps: the plant straws containing 10 to 25 percent of water are treated by steam explosion under the condition that the steam pressure is 0.9 to 1.9 MPa, and then the plant straws are air dried; with the synergistic effect of ferulic acid esterase and the degrading enzyme of lignocellulose material, the air dried plant straws processed by steam explosion is carried out zymohydrolysis to obtain the enzymolysis liquid of the ferulic acid. The steam explosion technology is clear and high efficient without pollution. Combined with enzymatic extraction, the method improves nine to twelve times of the release rate of the ferulic acid in the plant straws.

Description

Technical field: [0001] The invention belongs to the application field of biotechnology, and in particular relates to a method for improving the extraction rate of ferulic acid from plant stalks through steam explosion pretreatment. Background technique: [0002] Ferulic acid is an aromatic organic acid widely present in plant cell walls, and is one of the active ingredients of many Chinese herbal medicines such as Ferulicum and Angelica. It has been widely used in medicine, food, cosmetics and other industries. However, the content of ferulic acid in plant cell walls is relatively low, and most of them are combined between lignin and polysaccharides in the form of monomers or polymers, making the structure of cell walls more complex and dense, and ferulic acid is difficult to release. [0003] Ferulic acid is also widely found in crop straws, but it has not been developed and utilized on a large scale so far. One of the reasons is that the structure of plant straw cell wal...
