Embodiment Construction

[0015] The present invention induces and extracts the method for metallothionein from housefly larvae, and its operating steps are as follows:

[0016] a. Using wheat bran, water, 10% EM compound flora and a compound ion inducer containing zinc, copper and chromium as raw materials, ferment at room temperature for 10 to 50 hours to prepare a culture medium;

[0017] B, get the ovum of housefly and implant in the culture medium that step a makes, temperature is controlled at 30~35 degrees centigrade, after 24 hours, evenly spray 5% concentration on the surface of culture medium that is equivalent to 10% of the total amount of culture medium Cd 2+ , Zn 2+ 、Cu 2+ A metal ion inducer, cultivated for 4 to 6 days to obtain the protein material of housefly larvae;

[0018] c. Take the larvae cultured in step b, rinse them twice in pure water at 4-20 degrees Celsius, and then soak them in 10% hydrogen peroxide solution at 20-50 degrees Celsius for 10-120 minutes;

[0019] d. Rinse...

Abstract

The invention relates to a method for inducing and extracting metallothionein from housefly larva, which comprises the following steps of: culturing 3-5-day housefly larva in a nonmetal culture dish by using a culture medium of a metal ion biological inductive agent of Cd2+, Zn2+ and Cu2+; and carrying out rinsing, sterilizing, low temperature and high speed homogenate, membrane filtration, chromatographic fractionation, elution and concentration, and vacuum freezing and drying on the housefly larva to obtain Zn/Cu metallothionein which has molecular weight of 5300-6500 dDa and contains 20-27 percent of cysteine and subtypes thereof, wherein the molecular structure of the Zn/Cu metallothionein takes less aromatic amino acids. The yield of the metallothionein in biologically induced larva protein can reach 1mg/g. The production method has low cost, high purity and high yield of metallothionein.

Description

technical field [0001] The invention relates to a material extraction method, in particular to a method for inducing and extracting metallothionein from housefly larvae. Background technique [0002] Metallothionein is a metal-binding protein with a molecular weight of approximately 6000-8000 Da and is rich in cysteine. Metallothionein is widely distributed in prokaryotic and eukaryotic organisms, and has the functions of participating in the transport, storage and metabolism of rare elements, as well as detoxification of heavy metals and scavenging free radicals. The protein has been used to improve the microenvironment of the digestive tract, treat gastric ulcer and formulate cosmetics. There are many extraction and purification methods of metallothionein, including chromatography, high performance liquid chromatography, capillary electrophoresis, etc., but most of them can only be used for laboratory research and are not suitable for industrial production. And at presen...
