Embodiment 1

[0008] The materials used in this example are Porphyra phycoerythrin R-PE and throat cancer Hep-2 cell line, wherein Porphyria phycoerythrin R-PE was purchased from Hangzhou Aowei Bioengineering Co., Ltd., and throat cancer Hep-2 2 Cell lines were purchased from the Institute of Cells, Chinese Academy of Sciences.

[0009] (1) Throat cancer Hep-2 cell line was cultured with 1640 culture medium containing 10% calf serum, and the culture conditions were 37°C and 5% CO 2 concentration and saturation humidity;

[0010] (2), when the cells grow vigorously, digest the cells with 0.25% trypsin containing 0.02% ethylenediaminetetraacetic acid (EDTA), wash and blow off the cells and dilute to 10 6 / ml, 100 μL per well was inoculated in a 96-well plate;

[0011] (3), with a concentration of 100 μg / mL and a purity of 5.5 (A 564nm / A 280nm ) Serum-free culture medium of Porphyra phycoerythrin R-PE to replace the above-mentioned culture solution, cultivated for 4 hours;

Abstract

The invention discloses the application of phycobiliprotein in preparation of photosensitizer for treating oral and pharyngeal cancer cell by using an iodine-tungsten lamp. The phycobiliprotein of the invention is selected from porphyra yezoensis phycoerythrin R-PE and porphyra yezoensis phycocyanin C-PC extracted from porphyra yezoensis; under the light of the iodine-tungsten lamp, the effective photosensitizer has killing effect to the Hep-2 cells of the oral and pharyngeal cancer and high killing ability. The raw material is abundant in source and cheap in price; the extract process is simple; the iodine-tungsten lamp as the light source is small in volume and cheap in price and has low cost of the light source; in brief, the porphyra yezoensis phycoerythrin R-PE or the porphyra yezoensis phycocyanin C-PC as the photosensitizer kills the Hep-2 cells of the oral and pharyngeal cancer under the light of the iodine-tungsten lamp, greatly reduces the use cost in the course of the photodynamic therapy and is beneficial to wide popularization and use.

Description

Technical field: [0001] The invention relates to a medicine for clinical use in medical oncology, in particular to the application of a phycobiliprotein in the preparation of a photosensitizer for treating throat cancer cells with an iodine-tungsten lamp. Background technique: [0002] Photodynamic therapy is a new type of tumor therapy established and developed in the past 20 years. Its principle is mainly to achieve the killing effect on tumor cells by enriching the photosensitizer in the lesion and producing photosensitive damage under light. Photodynamic therapy depends largely on the nature of the photosensitizer employed. Traditional photosensitizers have serious skin phototoxicity and other shortcomings, and the development of new photosensitizers has become a research hotspot. Phycobiliprotein is a good pure natural pigment, which can be used in food, cosmetics, fluorescence detection and other fields. In recent years, according to the characteristics of its absorp...
