The invention discloses a fusion tandem antibacterial peptide and a preparation method thereof, relates to an antibacterial peptide and a preparation method thereof and solves problems that existing antibacterial peptide synthetic methods are low in yield and high in production cost and particularly the expression quantity is low or the expression is difficult caused by the fact that antibacterial peptide molecules are prone to be subjected to enzymolysis during the recombinant expression of the antibacterial peptide by genetic engineering. Four types of antibacterial peptides of fowlicidin-2, cecropin B, cecropin a (1-8)-melittin (1-18) and thanatin are in a series connection to form the fusion tandem antibacterial peptide. The preparation method includes (1) designing a cDNA fragment; (2) constructing a recombinant expression vector; (3) obtaining a transformant; and (4) inducing, collecting a supernatant and performing protein purification. The preparation method is used for the field of antibacterial peptide preparation.
