Embodiment 1

[0023] The screening method of the actinomycetes that the present invention has inhibitory effect to cucumber Phytophthora is as follows:

[0024] 1. Collection of soil samples

[0025] Four soil samples were randomly collected from near the Jinbian River in Zhangjiajie National Forest Park, Hunan Province. When collecting soil samples, first remove the topsoil, take about 200g of soil samples at a depth of 5-20cm, pack and mark them, and bring them back to the laboratory.

[0026] 2. Isolation of actinomycetes

[0027] Separation by dilution. Weigh 10 grams of the soil sample, put it into a conical flask equipped with small glass beads and 90 ml of sterile water, shake it for 20 minutes to dilute it. After standing for 30 seconds, dilute 10 times sequentially to prepare 10 -4 、10 -5 、10 -6 Dilution solution, draw 0.1ml of the dilution solution and add dropwise to Gao’s No. 1 medium (20 grams of soluble starch, 20 grams of agar, K 2 HPO 4 0.5g, NaCl 0.5g, KNO 3 1 g...

Embodiment 2

[0044] On February 10, 2012, cucumber seedlings were cultivated, and the variety was Shuyan Baiyu. Soak cucumber seeds in warm water at 50°C for 10 minutes, then overnight at 26°C. On February 11, the seeds were wrapped in gauze and kept at 26°C to accelerate germination. On February 12th, prepare seedling trays, sow seeds, and put them into a light incubator at 27°C for 12h during the day and 16°C for 12h at night, keeping RH80%. On February 29, the cucumber seedlings were inoculated with sporangia of Phytophthora cucumber when two true leaves appeared, and the concentration of sporangia was 5×10 3 zoospores / ml. Before inoculation, water the seedling tray thoroughly, and use the root irrigation inoculation method to take 2ml of inoculation suspension and add it around the roots of the plants. The inoculated cucumber seedlings were divided into 2 groups. One group was perfused with 2ml diluted 20×Gaoshi No. 1 culture solution in the roots, and the other group was perfused ...

Abstract

The invention relates to actinomycete capable of inhibiting phytophthoramelonis and a screening method of the actinomycete. Streptomyces purpeofuscus with preservation number CGMCC No.6191 is preserved in China general microbiological culture collection center. The streptomyces purpeofuscus is safe, nontoxic and effective antagonistic actinomycete capable of inhibiting phytophthoramelonis. The actinomycete has no pathogenicity to people and livestock, can be safely used for biological prevention and control of phytophthora melonis katsura and also has certain inbibitional effect on pyricularia grisea, phytophthora capsici leonian, phytophthora parasitica var nicotianae and colletotrichum glecosporioides. At present, few reports on phytophthoramelonis antagonistic bacteria are few in China, particularly reports on inhibiting action of the streptomyces purpeofuscus on phytophthoramelonis do not exist, and therefore, the antagonistic strain has a good application prospect.

Description

technical field [0001] The invention relates to a new bacterial strain capable of inhibiting Phytophthora cucumber and a screening method thereof. Background technique [0002] Cucumber Phytophthora Blight is caused by Cucumber Phytophthora ( Phytophthora melonis ) is a soil-borne disease caused by ferocious onset and rapid spread, causing large areas of cucumbers to die all the year round and having a great impact on cucumber production. The pathogen can not only harm cucumbers, but also infect pumpkins, loofahs, watermelons, gourds, melons, cantaloupes, bitter melons and other Cucurbitaceous plants. [0003] Cucumber blight can be diseased at any stage and in any part of the cucumber growth process. Pathogens can be transmitted through a variety of routes, mainly damaging fruits, leaves and stems, especially at the base of stems. Onset at the seedling stage, water-soaked soft rot or damping at the base of the stem, dark green or brown, softened and constricted, the uppe...
