The method comprises the following steps: (1) inoculating an enzyme production strain into a fermentation medium, and culturing for 30-80 hours so as to prepare a fermentation liquor; (2) carrying out thallus separating, thallus wall breaking and enzyme purifying on the fermentation liquor so as to prepare a trehalose synthase liquor; (3) carrying out a crosslinking embedding reaction so as to prepare the immobilized trehalose synthase; and (4) mixing the immobilized trehalose synthase with a maltose substrate solution, conversing and separating to obtain the trehalose.
