[0039] (1) Cultivation of ammonia oxidizing archaeal flora

[0040] Through previous research, it was found that a sewage treatment plant A 2 / O process (Anaerobic-Anoxic-Oxic) return sludge contains a large number of ammonia-oxidizing archaeal bacteria. Enrichment culture by replacement culture: Take a 1L Erlenmeyer flask and add 500mL enrichment medium (add 0.025gKH to 1L distilled water 2 PO 4 , 1.25gKHCO 3 , 7.5g casamino acids, 10g yeast extract, 3g sodium citrate, 2g KCl, 0.1gMgSO4 7H 2 O, 10g Na 2 CO 3 , adjust the pH to 7.2, and sterilize at 121°C, high temperature and high pressure for 30 minutes), and inoculate with 250mL activated sludge with ρ(SS) of 12200mg / L. Under the conditions of pH 6.5 and 140rpm / min, the cells were cultured on a constant temperature shaker at 30°C. When the number increased by an order of magnitude, the first-generation culture was terminated and replaced.

Abstract

The invention relates to an ammonia-oxidizing archaea flora cultivating method and application thereof in aerobic composting. The cultivating method comprises the following steps of: the first step, collecting sludge containing an ammonia-oxidizing archaea flora and preparing an activated sludge solution; the second step, performing enrichment culture in a substituting culture way, thereby obtaining a first-generation culture solution; the third step, under the conditions that the pH is 6.5, the rotation speed is 140 rpm / min and the constant temperature is 30 DEG C, performing shake cultivation on the first-generation culture solution, finishing the first-generation culture when the quantity is increased by an magnitude order, and then carrying out substitution; the four step, performing enrichment culture on the first-generation culture solution which is subjected to shake cultivation previously, thereby obtaining a second-generation culture solution; the fifth step, continuously repeating the shake cultivation to the fifth to eighth generation, thereby obtaining 800 mL of enriched ammonia-oxidizing archaea flora. The cultivating method is capable of remarkably increasing the quantity of the ammonia-oxidizing archaea, promoting the progress of a digestion reaction, reducing the emission of N2O and reducing the loss of nitrogen.

Description

technical field [0001] The invention relates to the technical field of microbes, in particular to a method for cultivating ammonia-oxidizing archaea flora and its application in aerobic composting. Background technique [0002] At present, aerobic composting technology, as an economical and efficient technology for the harmlessness, recycling and stabilization of organic solid waste, has been widely used in the treatment of livestock and poultry manure. However, the amount of nitrogen lost during composting is quite staggering. For example, in 2002, Barrington et al. summarized that when different groups of mixed wastes such as livestock and poultry manure-straw, sludge-straw, and livestock and poultry manure-sawdust were jointly composted, the nitrogen loss of each group of mixed wastes during the composting process was about Feed 16% to 76% of the total nitrogen. In 1992, the research of Martins et al. showed that the nitrogen loss caused by the volatilization of ammonia...
