Embodiment Construction

[0032] The specific embodiments of the present invention are given below with reference to the accompanying drawings, which are used to further describe the structure of the present invention.

[0033] The technical scheme of the present embodiment is as follows:

[0034] 1. Experimental part

[0035] Reagents and materials: All reagents are from Sinopharm Chemical Reagent Co., Ltd.

[0036] 1. Preparation of rhodamine hydrazide

[0037] Add rhodamine 6G (4.79g, 0.01mol), 5ml 80% hydrazine hydrate, and 30ml methanol into a 50ml flask, and reflux the mixed solution in an oil bath at 80-90°C for 4-5h. After cooling, filter to obtain a dark red crude product. After drying, Using DMC / methanol=10 / 1 silica gel column to separate and purify, the red product rhodamine hydrazide was obtained, the yield was 90%, and it was set aside.

[0038] 2. Rhodamine-hydrazone acetylpyridine fluorescent probe

[0039] Add rhodamine hydrazide (1.40g, 0.003mol), 2,6-diacetylpyridine (0.50g, 0.003...

Abstract

The invention relates to a rhodamine fluorescent probe for detecting Fe <2+>, and a preparation method thereof. The preparation method comprises the following steps: enabling rhodamine 6G and hydrazine hydrate to perform reflux reaction in alcohol to generate rhodamine hydrazide, then enabling rhodamine hydrazide and 2,6-diacetylpyridine to perform reflux reaction in alcohol, and through neutral aluminium trioxide column chromatography purification, obtaining the rhodamine acylhydrazone and acetyl pyridine fluorescent probe. The fluorescent probe is high in synthesis productivity, quick in reaction and high in selectivity, has high specificity to Fe <2+>, can work under the physiological environmental condition with the pH value of 7.4, and has a prospect of being applied to bioluminescence imaging.

Description

technical field [0001] The invention relates to a method for detecting ferrous ion Fe 2+ The rhodamine fluorescent probe and its preparation method belong to the technical field of fluorescent probes and its preparation method. Background technique [0002] Attention due to the increasingly important role of trace iron in biological and environmental factors [1-4] . The main function of iron in biology is to provide redox potential, which facilitates iron in many biochemical reactions in Fe 2+ and Fe 3+ transition between states [5] . High concentrations of iron ions in the body can increase the probability of certain diseases or tissue dysfunction. Disturbances in the body's iron levels also increase the incidence of certain disorders, such as metabolic anemia, liver and kidney damage, diabetes, heart failure, and neurodegenerative diseases [5-7] . Therefore, a simple and fast method for analyzing iron in biological samples will play an important role in biological ...
