Embodiment 1

[0035] Preparation and characterization of embodiment 1 nano-selenium

[0036] In this embodiment, three kinds of nano-selenium were designed and synthesized, which are respectively polyethylene glycol (PEG) dispersed nano-selenium (PEG-SeNPs) prepared by dissolving gray selenium by physical method, and utilizing vitamin C (VC) to reduce selenous acid (H 2 SeO 3 ) or sodium selenite (Na 2 SeO 3), and then use PEG or polyvinylpyrrolidone (PVP) to modify nano-selenium to obtain (PEGW-SeNPs, PVP-SeNPs), and detect their responsiveness to radiation, select the optimal material for further activity screening, and Study its co-action mechanism with X-rays.

[0037] (1) Preparation of PEG-SeNPs: reference [ZhengSY, LiXL, ZhangYB, XieQ, WongYS, ZhengWJ, et al. Specifically, under magnetic stirring, a certain amount of gray selenium is dispersed in PEG (ratio: 1:10-100), heated to 150-250°C, and maintained for 10-120min, the high-temperature system is rapidly cooled, and the obtain...

Embodiment 2

[0045] Example 2 Study on Anti-cervical Cancer Tumor Activity of PEG-SeNPs and X-ray in Vitro

[0046] Based on the electron-ray sensitivity characteristics of PEG-SeNPs, we look forward to developing the application of PEG-SeNPs in the field of radiotherapy. In this example, we studied the in vitro anti-tumor activity of three kinds of nano-selenium PEG-SeNPs, PEGW-SeNPs and PVP-SeNPs combined with radiotherapy. After Hela cells were cultured for 24 hours, different concentrations of nano-selenium were added for pretreatment for 6 hours, and then 8Gy of radiotherapy was applied on a clinical X-ray radiotherapy instrument. From Figure 4 A shows that the treatment groups of three kinds of nano-selenium alone found that the inhibition rates of PEG-SeNPs, PEGW-SeNPs and PVP-SeNPs on Hela cells were similar, and there was no significant difference. When co-treated with X-ray (8Gy), the cell inhibition rate of the combined treatment group of PEGW-SeNPs and PVP-SeNPs did not chan...

Embodiment 3

[0049] The influence of embodiment 3X-ray (8Gy) on PEG-SeNPs

[0050] From Figure 6 A shows that after 6 hours of treatment, PEG-SeNPs entered Hela cells and filled the cytoplasm, and from the results of nuclear staining, PEG-SeNPs were mainly distributed in the cytoplasm rather than the nucleus. Interestingly, after 8Gy of radiotherapy, the green fluorescence accumulated in Hela cells faded, while the blue color of the nucleus was still visible. We speculate that this is because PEG-SeNPs are sensitive to X-ray and then the intracellular disintegration and dispersion affect the intensity of green fluorescence. From Figure 6 The results of B show that after X-ray (8Gy) treatment, we also observed the morphology change of PEG-SeNPs.

[0051] We investigated the level of ROS produced by different nanoselenium treated with radiotherapy and PEG-SeNPs into Hela cells. From Figure 7 It can be seen that the level of ROS produced by PEG-SeNPs in the cell-free model is higher, ...

Abstract

The invention discloses an application of nano selenium as an X-ray radiation therapy sensitizer. In the invention, elementary selenium is dissolved in PEG to prepare the nano selenium (SeNPs), nano selenium modified by polysaccharide and chitosan and tumor-targeted nano selenium modified by folic acid and transferrin. The SeNPs has an X-ray stimulating and responding character and has a synergistic effect with the X-ray, thereby high-effectively inhibiting proliferations of human cervical carcinoma cell Hela, melanoma cell A375, osteosarcoma cell MG-63, breast cancer cell MCF-7 and brain glioma cell C6 and the like. The main mechanism of the SeNPs is that by means of increase of active oxygen accumulation in cells, further cell period retardant tumor cell apoptosis is induced. A result proves that the SeNPs can be developed as a novel radiation therapy sensitizer.

Description

technical field [0001] The invention relates to the technical field of tumor treatment, in particular to the application of nano-selenium as an X-ray radiotherapy sensitizer. Background technique [0002] In the past ten years, radiotherapy has developed rapidly and is widely used in clinical tumor treatment. Studies have shown that about 50% of cancer patients have undergone radiotherapy. However, little is known about the mechanisms by which radiotherapy inhibits tumor growth. What's more, radiotherapy can easily lead to tumor cell resistance to radiotherapy, which hinders the treatment of the disease. Cervical cancer is a malignant tumor that occurs frequently in women. In the treatment of cervical cancer, the combination of chemotherapy and radiotherapy, that is, radiotherapy and chemotherapy, is often used for treatment. Unfortunately, cervical cancer is prone to resistance to both chemotherapy drugs and radiation therapy. [0003] The side effects caused by chemoth...
