Abstract

The invention discloses a method for inducing production of Rosa sterilis tetraploid through soaking method. The method comprises a first step of preparation of material: removing leaves and tops of aseptic seedling stems; cutting the aseptic seedling stems into 0.8 to 3 cm in length; remaining at least one axillae for each section; a second step of pre-cultivation: pre-cultivating the stems in a culture medium MS+ 0.5mg / L 6-BA for 2 to 3 days; a third step of vibration in a manner of avoiding light: soaking the pre-treated material into a sterile water containing 200 to 500mg / L of colchicine; vibrating the material on a shaking table at 120r / min and 25 to 30 DEG C for 12 to 72 hours in a manner of avoiding light; a fourth step of light cultivation: after being soaked, transferring the material into a culture medium MS+ 0.15mg / L NAA+ 0.5mg / L 6-BA without containing colchicine to be cultivated continuously; a fifth step of homogenization treatment for variant buds: after the cultivation in the fourth step is performed for 40 days, selecting the buds which are obviously variant, repeatedly cultivating the buds in the culture medium MS+ 0.15mg / L NAA+ 0.5mg / L 6-BA five times at interval of 40 days.
