Problems solved by technology

Although liver biopsy is the gold standard for judging the degree of liver fibrosis, the invasiveness of the sampling process makes the follow-up of patients poor
In recent years, the emerging hepatic elasticity measurement technology has been gradually applied in clinical practice, but the equipment is expensive and difficult to apply in field research. Therefore, the development of non-invasive, simple, efficient and accurate diagnostic methods is particularly urgent

[0030] To establish a mouse model of Schistosoma japonicum infection: According to the conventional method, take the positive snails and place them in dechlorinated water under the light condition of 25°C-28°C to release cercariae for 1-2h. Dip the water surface cercariae with an inoculation loop and collect them on the coverslip. Clean grade 8-week-old BALB / c mice were anesthetized with pentobarbital, and fixed on the mouse board with their backs up. After plucking the abdomen, clean and moisten the abdomen of the mice with dechlorinated water overnight, dip 15 cercariae on the water surface with an inoculation loop and put them on the cover glass, and attach them to the abdominal skin of the mice for infection. According to the length of infection time, they were divided ...

Abstract

The invention discloses the application of human β-actin as a serological diagnostic marker for the occurrence of schistosomiasis hepatic fibrosis, and the amino acid sequence of the β-actin protein is shown in SEQ ID NO:1. The study found that in the liver of Schistosoma japonicum hepatic fibrosis mice and patient serum, the gene and protein levels of β-actin were unstable, and its content in the liver increased significantly with the development of fibrosis, and the content in serum increased with the development of fibrosis. The development of fibrosis was significantly reduced. Therefore, the expression level of β-actin has become an important diagnostic index for judging the occurrence of liver fibrosis and the degree of lesion in patients with schistosomiasis japonicum. By detecting the expression level of β-actin in peripheral blood of patients, it can provide more information for field investigation and clinical work. It is a simple, easy-to-operate, economical and scientific diagnostic technique for liver fibrosis in Schistosomiasis japonicum.

Description

technical field [0001] The invention relates to the technical field of detection of hepatic fibrosis in schistosomiasis japonicum, in particular to the application of β-actin protein as a serological diagnostic marker for the occurrence of hepatic fibrosis in schistosomiasis japonicum. Background technique [0002] Schistosoma japonicum ( Schistosoma japonicum ) infection caused by Schistosomiasis japonicum is a zoonotic parasitic disease that is widespread and seriously endangers human health and social and economic development. Globally, more than 200 million people suffer from the disease, resulting in a loss of 1.53 million disability-adjusted life years. my country is one of the countries where the prevalence of schistosomiasis is the most serious. It has carried out comprehensive and planned control work for more than half a century and has achieved remarkable results. According to statistics, by the end of 2013, there were still 184,000 schistosomiasis patients in m...
