Embodiment 1

[0033] The invention provides a kind of rapid breeding method using Chinese cabbage axillary buds as explants, specifically comprising the following steps:

[0034] 1) Select Chinese cabbage at the heading stage, and peel off the outer leaves of the leaf bulb. Cut off the axillary buds at the junction of the base of the petiole and the shortened stem with a scalpel, as figure 1 As shown, the incision is located about 3 mm below the base of the axillary bud and contains part of the shortened stem tissue.

[0035] 2) Under aseptic conditions, first disinfect the surface with 70% alcohol by volume for 30 seconds, and then rinse with sterile distilled water. Then pour 5% NaClO solution to sterilize for 15 minutes, rinse with sterile distilled water 5-6 times, 3 minutes each time. After rinsing, use sterilized absorbent paper to dry the excess water absorbed by the axillary buds. After sterilization, excess shortened stem tissue at the base of the axillary buds is excised.

Embodiment 2

[0043] The present embodiment compares the influence of NaClO solution disinfection of different mass concentrations on the growth of explants at different times by experiment, and the specific experimental steps are as follows:

[0044] 1) Select 'Xibai No. 4' Chinese cabbage at the heading stage, and peel off the outer leaves of the bulb. The axillary bud at the junction of the base of the petiole and the shortened stem was cut off with a scalpel. The cut was located about 3 mm below the base of the axillary bud, and contained part of the shortened stem tissue.

[0045] 2) Under aseptic conditions, first disinfect the surface with 70% alcohol by volume for 30 seconds, and then rinse with sterile distilled water. Pour NaClO solutions of different mass concentrations to sterilize for different times (as shown in Table 1), and rinse with sterile distilled water 5-6 times, each time for 3 minutes. After rinsing, the excess water absorbed by the axillary buds was blotted dry wit...

Embodiment 3

[0056] In this embodiment, the influence of different TDZ (Thidiazuron, thiadiazole phenylurea) and NAA (1-naphthylacetic acid, α-naphthalene acetic acid) concentration ratios on the regeneration of Chinese cabbage axillary buds is compared through experiments. The specific experimental steps are as follows:

[0057] 1) Select 'Xibai No. 4' and '87-114' Chinese cabbage at the heading stage, and peel off the outer leaves of the bulb. The axillary bud at the junction of the base of the petiole and the shortened stem was cut off with a scalpel. The cut was located about 3 mm below the base of the axillary bud, and contained part of the shortened stem tissue.

[0058] 2) Under aseptic conditions, first disinfect the surface with 70% alcohol by volume for 30 seconds, and then rinse with sterile distilled water. Then pour 5% NaClO solution to sterilize for 10 minutes, rinse with sterile distilled water 5-6 times, 3 minutes each time. After rinsing, use sterilized absorbent paper to...

Abstract

The invention provides a rapid propagation method by taking Chinese cabbage axillary buds as explants. The rapid propagation method specifically comprises the following steps: cutting off axillary buds as explants; sterilizing and cleaning the axillary buds; culturing the axillary buds on a sprout enrichment culture medium; cutting off buds enriched on the explants, dipping an IBA (Indolebutyric Acid) solution, and performing adventitious root induction in a non-agar solid medium; and transferring regeneration seedlings into a flowerpot substrate to be arranged at room temperature, and performing natural light culture. According to the method disclosed by the invention, the Chinese cabbage axillary buds are taken as the explants, and the use concentration of plant growth regulating substances in the enrichment medium is optimized. Individual enrichment is performed by utilizing existing bud primordium, the regeneration efficiency of Chinese cabbage tissue culture is improved, the bud formation speed is high, and the influence of genotype is slight. The culture process is simple, excellent breeding materials and germplasm resources can be subjected to timely expanding propagation and utilization in a short time, the breeding process is greatly accelerated, and a novel pathway is provided for solving the regeneration problem of AA genomic crops.

Description

technical field [0001] The invention relates to the technical field of plant tissue culture, in particular to a rapid breeding method using Chinese cabbage axillary buds as explants. Background technique [0002] Chinese cabbage (Brassica rapa L.), which originated in China, has always been one of the most important vegetables in Asia due to its wide planting area and huge consumption. With the improvement of living standards and vegetable production levels, consumers have more and more urgent demands for the diversification of Chinese cabbage varieties (Zhang Fenglan et al., 2017). Improving breeding efficiency is the key to speeding up the breeding of new varieties of Chinese cabbage. As an important biotechnology tool, tissue culture plays an irreplaceable role in the rapid propagation of various materials (excellent mutants, inbred lines, hybrid combinations and landraces, etc.) in the breeding process of Chinese cabbage. So far, researchers at home and abroad have est...
