Abstract

The invention provides a method for inductively differentiating adipose derived mesenchymal stem cells into vascular endothelial cells. The method comprises the following steps: washing adipose tissues; adding a tissue digestive enzyme which has an equal volume with the adipose tissues for digestion; after digestion is finished, adding trypsin liquid which has the equal volume with the tissue digestive enzyme for digestion; after the digestion is finished, centrifuging to remove upper-layer oil and digestion liquid, wherein sediment is adipose tissue stromal vascular fraction cells; carrying out re-suspension on the stromal vascular fraction cells again through normal saline and sieving to remove large tissues; washing with the normal saline again and centrifuging to remove supernatant; inoculating and culturing washed stromal vascular cells through a serum-free multiplication culture medium, so as to obtain the adipose derived mesenchymal stem cells; selecting P3-generation adipose derived mesenchymal stem cells and digesting to obtain single-cell suspension; inoculating and culturing overnight by utilizing the serum-free multiplication culture medium; after culturing for 24 h, removing the serum-free multiplication culture medium; and adding an induction culture solution and carrying out induced culture, so as to obtain the differentiated vascular endothelial cells.
