Embodiment 1

[0038] A preparation method for delivery of microRNA nanocarrier based on zeolite imidazole skeleton, comprising the steps of:

[0039] (1) dissolving miR-34a in sterile, enzyme-free water (appropriate amount of sterile enzyme-free, dissolving miR-34a) to obtain miR-34a aqueous solution;

[0040] (2) The miR-34a aqueous solution is mixed with ZIF-8 nanoparticles, so that the mass ratio of miR-34a and ZIF-8 nanoparticles is 1:10, and the microRNA nanocarrier based on the zeolite imidazole skeleton is obtained, referred to as ZIF- 8 / miR-34a. See figure 1 A Transmission electron microscope image.

Embodiment 2

[0042] A preparation method for delivery of microRNA nanocarrier based on zeolite imidazole skeleton, comprising the steps of:

[0043] (1) dissolving miR-34a in sterile, enzyme-free water (appropriate amount of sterile enzyme-free, dissolving miR-34a) to obtain miR-34a aqueous solution;

[0044] (2) The miR-34a aqueous solution is mixed with ZIF-8 nanoparticles, so that the mass ratio of miR-34a and ZIF-8 nanoparticles is 1:55, and the microRNA nanocarrier based on the zeolite imidazole skeleton is obtained.

Embodiment 3

[0046] A preparation method for delivery of microRNA nanocarrier based on zeolite imidazole skeleton, comprising the steps of:

[0047] (1) dissolving miR-122 in sterile enzyme-free water (sterile enzyme-free appropriate amount, dissolve miR-122) to obtain miR-122 aqueous solution;

[0048] (2) The miR-122 aqueous solution is mixed with ZIF-8 nanoparticles, so that the mass ratio of miR-122 and ZIF-8 nanoparticles is 1:100, and the delivery microRNA nanocarrier based on the zeolite imidazole skeleton is obtained.

[0049]The transmission electron micrographs of the delivery microRNA nanocarrier based on the zeolite imidazole skeleton in Example 2 and Example 3 are similar to the microRNA nanocarrier delivery based on the zeolite imidazole skeleton in Example 1.

Abstract

The invention discloses a microRNA delivery nanocarrier based on a zeolitic imidazolate framework and a preparation method thereof. The preparation method comprises the steps of (1) dissolving microRNA in aseptic enzyme-free water to obtain a microRNA solution; and (2) mixing the microRNA solution with ZIF-8 (zeolitic imidazolate framework 8) nanoparticles to obtain the microRNA delivery nanocarrier based on the zeolitic imidazolate framework. The preparation method is simple, has high loading efficiency, and is simple and low in cost. The microRNA delivery nanocarrier based on the zeolitic imidazolate framework can effectively protect the microRNA from being degraded. The ZIF-8 can split in a weak acidity environment to accelerate the rapid release of the microRNA at tumor cells, therebyenhancing therapeutic effect of drugs. The microRNA delivery nanocarrier based on the zeolitic imidazolate framework prepared herein has good biocompatibility.

Description

technical field [0001] The invention belongs to the field of biomedical materials, and relates to a delivery microRNA nanometer carrier based on a zeolite imidazole skeleton and a preparation method. Background technique [0002] Cancer threatens human health. Currently, the main means of cancer treatment are: surgery, radiotherapy, drug chemotherapy and immunotherapy. Among them, drug chemotherapy is the main treatment method, but drug chemotherapy has many side effects, such as poor targeting, low drug utilization and other defects. [0003] Therefore, there is a need to develop nanocarriers, and the main purpose of using nanocarriers for drug delivery is to improve pharmacokinetics and alter the biodistribution of drugs. Thereby enhancing therapeutic efficacy and reducing side effects by preferentially accumulating drugs in diseased tissues. The nanocarriers include inorganic nanomaterials such as silicon dioxide, quantum dots, and metal nanoparticles, and organic nanom...
