The method comprises thesteps as follows: performing extraction on hibiseu manihot flowers by a microwave assisted ethanol aqueous solution, obtaining petroleum ether phase, ethyl acetate, n-butyl alcohol and aqueous phaseextracts, and realizing primary purification by use of different solubility of the 6 flavonoids of the hibiseu manihot flowers in each extract phase; and separating the mixture by polyamide rapid preparative chromatography and preparative high performance liquid chromatography to obtain the 6 flavonoid compounds including the rutin, the hyperoside, the isoquercitrin, the hibifolin, the myricetin and the quercetin with the purity higher than 95.0%. The method is simple to operate and shorter in production cycle and can be used as a preferred method for separating and purifying high-purity flavonoids from the hibiseu manihot flowers, and the prepared products have high purity.
