[0039] This embodiment provides a purification process of highland barley β-glucan, including the following steps: using AB-8 macroporous resin to adsorb highland barley extract to remove pigment, the resin dosage is 2%-4%, the temperature is 40°C-60°C, pH2.0-4.0, time 70-90min. Centrifuge at 6000 r / min for 15 min to remove the resin in the solution and collect the supernatant. The supernatant was properly concentrated, passed through Sepharose CL-4B agarose column, polysaccharide was detected by anthrone-sulfuric acid method at 620 nm separator, and β-glucan was detected by Congo red method. Abstract

The invention discloses highland barley beta-glucan and a purification process thereof. The purification process comprises the steps of macroporous resin decoloration, agarose gel column chromatography molecular sieve purification, drying and the like. According to the invention, a new highland barley beta-glucan is separated from highland barley; the beta-glucan is subjected to structural identification by adopting a plurality of methods taking high performance gel chromatography-differential-laser scattering detection (HPSEC-dRI-LS) and a nuclear magnetic resonance carbon spectrum (13C-NMR)as main methods; through an enzyme inhibition experiment, the hypoglycemic activity of the highland barley beta-glucan which is purified by the method and has a definite structure and sequence is verified, and a foundation is laid for developing functional foods for regulating blood sugar.
