Embodiment Construction

[0019] Cases of sudden cattle death occurred in many places in Tongliao City in 2010, and one of the farmers had sudden death of cattle. After we checked the dead cattle for infectious diseases such as anthrax bacillus and foot-and-mouth disease virus, we conducted an autopsy and isolated bacteria from the liver and spleen. The bacteria were isolated and identified, and finally identified as Bacillus amyloliquefaciens. Before this, there was no relevant report on the death of this bacterial infection, so it was biologically identified.

[0020] Use the viscera of dead cows to touch the plate on ordinary agar medium, culture at 37°C for 12 hours, colonies appear on the medium, pick the colonies for staining, microscopic examination shows that the bacteria are Gram-positive bacteria with long rods shape, mostly in the form of two short chains ( figure 1 ).

[0021] The 16s rDNA sequence analysis of the bacteria was carried out: the strain was inoculated in LB liquid medium, 3...

Abstract

The invention relates to a livestock microorganism and an application thereof, in particular to bovine-derived bacillus amyloliquefaciens with epidemic pathogenicity and lethality and an application thereof. The bovine-derived bacillus amyloliquefaciens is preserved in the China General Microbiological Culture Collection Center (CGMCC) on April 25, 2019, and has a preservation number of CGMCC No.17627. The bacillus amyloliquefaciens is named in a classified manner. Strain information is as follows: the strain is separated from the liver and spleen of a cow suffering from the sudden death disease; under a bacterial colony staining microscope, the strain is in a long rod shape, mostly exists in a two-section short chain form, and is a Gram-positive bacterium; a 16SrDNA sequence is as shown in a sequence table; the strain is sensitive to ceftriaxone sodium, sulfamethoxazole and doxycycline, has relatively poor sensitivity to amoxicillin and is not sensitive to cefepime; the strain has relatively strong alkali resistance and salt resistance; the strain has relatively strong high temperature resistance; and animal experiments prove that the strain has the pathogenicity and lethality. The bacillus amyloliquefaciens is used for bovine epidemic disease quarantine control samples, and can also be used as a strain for bovine epidemic disease prevention vaccines.

Description

technical field [0001] The invention relates to a livestock microorganism and its use, namely a lethal bovine-derived bacillus amyloliquefaciens and its use. Background technique [0002] Bacillus amyloliquefaciens (Bacillus amyloliquefaciens) has always been considered as a probiotic for animals and plants, which can inhibit the growth of other harmful bacteria for animals and plants. However, in 2018, sudden death of cattle occurred in many places in Tongliao City, Inner Mongolia Autonomous Region. After screening the dead cattle for infectious diseases such as anthrax bacillus and foot-and-mouth disease virus, they were autopsyed and isolated from the liver and spleen. a type of bacteria. The bacteria were isolated and identified as Bacillus amyloliquefaciens. In-depth research on the pathogenicity and control measures of Bacillus amyloliquefaciens is of great significance for the development of cattle industry and the reduction of disease losses.
