Abstract

The invention discloses a preparation method of a biodegradable PLGA-SS-PEG nano-drug carrier. The preparation method comprises synthesis of a PLGA-SS-PEG high-molecular polymer and preparation of PLGA nano-particles; and the PLGA nano-particles with good properties are obtained through a centrifugal method. The invention further discloses a preparation method of a chemotherapeutic drug, traditional Chinese medicine monomer or gene drug loaded targeted molecule modified intelligent response PLGA nano-drug, and application of the PLGA nano-drug in preparation of drugs for treating lung cancer,liver cancer, brain cancer or pancreatic cancer. The nano-drug can enter tumour cells through targeting and endocytosis; and the carried anti-cancer drug can be controllably released under the condition of high content of glutathione in the tumour cells, so that the purpose of inhibiting the tumour cells more efficiently with low toxicity is achieved. The diameter of the nano-drug is about 110-500nm; the blank nano-particles have excellent biocompatibility; targeted modification can achieve the purpose of specifically conveying the nano-drug to tumour cells; and the carried anti-cancer drug can be controllably released under the condition of 5-9 mM reducibility GSH.
