Abstract

The invention provides a method for preventing dendrobium officinale tissue culture seedlings from being polluted by bacteria. The method is characterized by comprising the following steps of (1) preparing a nano-copper mother solution; (2) diluting the nano-copper mother solution; (3) preparing a disinfectant; (4) preparing a mixed culture medium I; (5) disinfecting dendrobium officinale seeds; (6), culturing the dendrobium officinale seeds; (7), performing differentiation culture on the dendrobium officinale seeds; (8), preparing a mixed culture medium II; (9), culturing differentiation immature embryos; and (10), culturing and transplanting bottle seedlings. The nano-copper solution with the concentration of 10<-2>% is added into the culture media, infection bacteria are blocked in theseed germination culture process and the immature embryo differentiation culture process, the bacterial contamination rate of dendrobium officinale is effectively reduced, the unique technical effectis achieved, and the blocking rate is as high as 93%. Damage of antibiotics to plants can be reduced, the survival rate of dendrobium officinale seedlings can be increased, even if pollution exists, blocking can be achieved, and therefore a large number of experimental materials can be preserved.

Description

technical field [0001] The invention relates to the field of new materials and plant tissue culture, in particular to a method for blocking bacterial contamination of dendrobium candidum tissue culture seedlings by using nanometer copper. Background technique [0002] Nanomaterials with one-dimensional, two-dimensional or three-dimensional dimensions of only a few nanometers or tens of nanometers are made by advanced physical, chemical, biological and other technologies, and their structures are very different from ordinary materials, such as nanograin boundaries. The atomic spacing is large, the density is low, and the atomic arrangement is random. In recent years, related research on the use of nano-copper for antibacterial has gradually attracted attention. Due to its quantum effect, small size effect, and large surface area, nano-copper exhibits better safety, antibacterial properties, and long-term antibacterial properties. [0003] The exposed seeds after pod crackin...
