Abstract

The method comprises the following steps: by taking DNA (Deoxyribose Nucleic Acid) of a to-be-detected fruit fly genome as a template, carrying out PCR (Polymerase Chain Reaction) amplification on an mtCOI gene segment: respectively carrying out enzyme digestion on a PCR product by adopting restriction endonuclease SapI, BsmI and / or Sac I; and judging the types of the fruit flies through gel electrophoresis. According to the invention, the COI gene sequence has sequence difference among different fruit flies, then restriction enzyme is utilized to recognize specific enzyme cutting sites, COI gene segments of three fruit flies can be cut into segments with different sizes, and finally, the three fruit flies can be quickly recognized and identified through agarose gel electrophoresis.
